Objective The expression of Mre11mRNA in parotid and submandibular gland cells of rat after irradiation by OGy,3Gy,6Gy,9Gy,12Gy,15Gy different doses, Compared with the unirradiated group, To observe the changes of Mre11gene and protein expression after irradiation on the different types of salivary gland epithelial cells, Detecte localization of Mre11in the normal different types of salivary gland epithelial cells, And then from the gene and protein levels of rat salivary gland cells after irradiation to investigating Mre11gene and protein expression changed.Methods Our studies were studied with60wistar rats, Use completely random number method to divided them into six groups by single irradiation of OGy,3Gy,6Gy,9Gy,12Gy,15Gy. Each group has10rats, then made the salivary glands due to exposure to ionizing radiation, each group received a dose ofOGy,3Gy,6Gy,9Gy,12Gy,15Gy respectively. After2hours, collect the salivary glands. Through transmission electron microscope to examine the effect of ionizing radiation on rat parotid gland. Expression of Mre11mRNA was detected through RT-PCR. Conduct Mre11gene expression level of semi-quantitative analysis by Mre11/β-actin, Mre11protein expression was assayed with immunocytochemistry. Use Image-Pro Plus5.0software to measure the average optical density (mOD) of each group. Results The semi-quantitative OD value of MrellmRNA in salivary gland of rat afterOGy,3Gy,6Gy,9Gy,12Gy,15Gy irradiation respectively. The One-way analysis of variance shows P=0.776(P>0.05) and P=0.954(P>0.05), the expression of Mrell mRNA had not statistieally significant between each group.We used immunofluorescence assay to detect Mrell protein of salivary gland cells after irradiation by OGy,3Gy,6Gy,9Gy,12Gy,15Gy. The One-way analysis of variance shows P<0.05, the expression of Mrell protein had statistieally significant. Fluorescence intensity first increased and then deereased. Through TEM, with the increse of dose, the irradiated rats had been destroyed. The damage became more and more serious and there was no recovery.Conclusion60Coy irradiation can lead damage to salivary glands, and the level of damage had much to do with radiation exposure dose, and the damage is inreversible. Mrell Protein expression with time first inereased and then decreased, and it is stronger in parotid gland cells than that in submandibular gland cells. The expression of Mrell mRNA in salivary gland cells of rat was independent of60Coy-ray dose escalation.
