Objective To investigate the effect of curcumin on the inhibition of TLR4/NF-κB signaling pathway and the expression of IL-23in ulcerative colitis rats and further to explore the mechanism of curcumin treating IBD.Methods Eighty SD rats were randomly divided into normal control gro-up(group A, n=20), TNBS-induced colitis group(group B, n=20), curcumin-tre-ated group (group C,n=20) and SASP-treated group (group D, n=20). Rat colitis model(group B, C and D)was established with100mg/kg body weight of TNBS enema, group A was treated with100mg/kg body weight of0.9%sodium chlori-de solution enema. Group A and B fed on100mg/kg body weight of0.9%sodiu-m chloride solution, group C fed on100mg/kg body weight of curcumin, group D fed on100mg/kg body weight of SASP.Colon intestinal samples was collect-ed after8days. Evaluated the weight changes of rats in each group, and rat inte-stinal pathological features and pathological scores was recorded, and evaluated disease activity index (DAI), the colonic mucosa damage index(CMDI) and hist-ological (HS). Determined myeloperoxidase (MPO) levels by immunohistoche-mical method; TLR4, NF-κB and IL-23of colon tissue was investigated by RT-PCR and western blot.Results Compared with the TNBS-induced colitis group, the scores of DAI, CMDI, HS, the expression of NF-κB and IL-23protein and mRNA were decreased significantly in colon mucosa of curcumin group (P<0.05).The expression of TLR-4protein was decreased significantly in curcumin group (P<0.05), the level of TLR-4mRNA was not changed among the groups (P>0.05).There was no significantl difference in TLR4,NF-κB and IL-23between curcumin treated group and SASP treated group (P>0.05). The results also showed that the scores of DAI was decreased rapid in curcumin treated group than SASP treated group on the third day (P<0.05).Conclusions Curcumin significantly inhibites IL-23expression in experimental colitis rats through inhibiting TLR4/NF-κB signaling pathway, and has significant anti inflammatory effect. Moreover,it is superior to SASP. |
