Objective To detect the drug sensitivity of human ovarian tumor stem cells to cisplatinand salinomycin and compare the effect of the two drugs to ovarian tumor stem cells.Methods Isolating and identifying ovarian cancer stem cells from SKOV-3cells byculturing them in serum free medium(SFM).Flow cytometry analysis were used toexamine the cell surface marker CD44.The IC50median inhibitory concentration ofsalinomycin and cisplatin was tested by cell counting kit-8(CCK-8)assay atfer48hours.Choose the IC50of the48th hour as the appropriate dosage.ABCBl and ABCBG2mRNAwere tested by RT-PCR in the experiment group and control group at48hour.Results CD44antigen was significantly higher in ovarian stem cells.The IC50ofsalinomycin and Cisplatin to ovarian cancer cell SKOV-3is(62.50士2.53),(3.62士0,38)u mol/L.The IC50of salinomycin and Cisplatin to ovarian cancer stem cell is(48.81±2.83),(3.80士0.22)ii mol/L in48th hour.The drug resistance index(RI)ofCisplatin is2.12±0.41,which is much higher than salinomycin(0.78士0.08)(t=6.22?p<0.05).The transcriptional level of ABCBl and ABCBG2of the ovarian cancer stem cellsatfer culturing in the appropriate dosage of salinomycin for48hours was lower in thecontrol group(t=7.49,9.75;P<0.05).Conclusion The ovarian cancer stem cell was more sensitive to salinomycin thancisplatin.The effect of Salinomycin for human ovarian cancer stem cells is stronger thanCisplatin.This results may be in connection with the high expression of ABCBl andABCG2genes in cancer stem cells.
