Relationship Of GIn223Arg Polymorphism In The Leptin Receptor To Cerebral Infarction And Abdominal Obesity | | Posted on:2013-12-12 | Degree:Master | Type:Thesis | | Country:China | Candidate:M Han | Full Text:PDF | | GTID:2234330371473029 | Subject:Neurology | | Abstract/Summary: | | | PURPOSE:To explore the Gln223Arg polymorphism in the leptin receptor(LEPR)and to analyze its correlation with cerebral infarction(CI)and abdominal obesity in Chinese Han people of Qingdao city.METHODS:The Gln223Arg polymorphism was studied by polymerase chain reaction and restrictive fragment length polymorphism(PCR-RFLP)in295patients with CI and256controls of healthy people.The two groups were respectively divided into subgroups with and without abdominal obesity on basis of abdominal circumference.The relationship of Gln223Arg polymorphism to CI and abdominal obesity was analyzed.The serum levels of leptin were measured in all the groups with the method of enzyme-linked immuno sorbent assay(ELISA).RESULTS:(1)There were significant differences in the distribution of genotype and allele frequencies of Gln223Arg polymorphism in LEPR between case group and controls.Compared to control group,case group had higher frequencies of genotype GG(19.32%VS10.55%,P=0.004).G allele frequency in case group were significantly higher than those in control group(46.61%VS30.86%,P<0.001).Making use of logistic regression,we detected carriers of G allele had an increased risk of CI,compared to subjects with A allele(OR(G/A)=5.687,95.0%CI:2.270~14.252,P<0.001).(2)No significant difference in the genotype distribution and allele frequencies of Gln223Arg polymorphism in people with and without abdominal obesity was observed(P=0.107,P=0.064).The serum levels of leptin in case group were higher than the control group(p<0.001).(3)However,the difference of the serum levels of leptin in people with and without abdominal obesity was of no statistical significance(p=0.870).CONCLUSION:Results from this study suggests that Gln223Arg polymorphism in LEPR is possibly associated with CI and not associated with abdominal obesity,and G allele is possibly associated with an increased risk of CI. | | Keywords/Search Tags: | leptin receptor, gene polymorphism, cerebral infarction, abdominalobesity, susceptibility | | Related items |
