Objective:To detect the serum EBV VCA-IgA and EA-IgA and peripheral blood mononuclear cells of EBV DNA load ofnasopharyngealcancer(NPC) patients before and after radiotherapy and discuss the significance of serum EBV DNA load and antibody tilter to NPC efficacy monitoring and prognostic evaluation.Methods:EBERl was detected in24NPC patients by in situ hybridization (ISH).The peripheral blood mononuclear cell samples were collected before, during and after radiotherapy. Enzyme linked immunosorbent assay (ELISA) and real-time PCR were taken to detect serum EBV VCA-IgA and EA-IgA and peripheral blood mononuclear cells of EBV DNA. EBV-specific antibodies and EBV DNA load changes with radiotherapy were analyzed. Results:The changes of VCA-IgA, EA-IgA and EBV DNA from22patients in three different stages of radiotherapy had significant differences(F=3.305,8.914,9.461, P<0.05). VCA-IgA after the radiotherapy was much lower than that of before the treatment (t=2.081,P<0.05), but there was no significant difference during the radiotherapy compared with that before or with those after the treatment (t=0.33,0.47; P>0.05). EA-IgA after the radiotherapy was much lower than that of before the treatment, but there was no significant differences during the radiotl erapy compared with that before or with those after the treatment (t=0.606,0.50, P<0.05).③EBV DNA load was detected in15NPC patients except7patients.EBV DNA before radiotherapy compared with during or after the treatment had significant difference (t=2.47,4.34; P<0.05), however, EBV DNA during the radiotherapy had no differences with that after the treatment(t=1.86, P>0.05).Conclusion:To dynamic observation the change of EBV VCA-IgA, EA-IgA antibody titer and EBV DNA load in peripheral blood has practical significance, and EBV DNA load change compared to the VCA-IgA and EA-IgA is more sensitive.②EBV DNA change observation in monitoring NPC treatment effect is prior to VCA-IgA and EA-IgA antibody titer detection. The combination detection and comprehensive analysis of EBV DNA load and antibody titer is helpful for assessment of the radiotherapy effect and prognosis of NPC patients.
