ObjectiveEndotoxemia is a high mortality disease, which induced by infection, serious burn or wound, can lead to septic shock, DIC, MODS, MOF, even lead to death. But there are currently no effective and safety anti-sepsis drugs in clinical use. HJM is a Chinese herbal preparation, which composed of scutellaria baicalensis, honeysuckle, moutan bark, and so on. Modern pharmacology study suggests that substance contains baicalin, chlorogenic acid, dan skin phenol have effect of anti-interference, antithrombotic and immunoregulation. In this study, we observed effect of HJM on endotoxemia mice and further investigated the mechanisms of its actions, in the hope of identifying a clinically relevant anti-sepsis drug.MethodsThis study included three parts:Firstly, we used "Koushi method" for E.coli endotoxin LD50 determination test. Secondly, we examined the effects of HJM on the mortality, clinical symptoms and organ histological changes of endotoxemia. mice Lastly, we investigated serum TNF-a, IL-1, IL-6, IL-4 and PGE2 levels by ELISA at 6h, 12h,18h,24h,48h and 72h after injection. Liver NO content and SOD activity were measured at the same time.Results1 Half an hour later after injection, the fur of endotoxemia mice were fleeciness, and their spirit withering, eyes were closed, cluster, the activity significantly reduced, and no eating.8 hours after injection, one of the mice in the high dosage group was dead, and dissection found much organ damage. The LD50 of E.coli endotoxin was 15.479 mL/kg,95% confidence limit was 13.452-17.783 mL/kg.2 The mortality in HJM treated groups were remarkably reduced, especially treatment with HJM at a dosage of 0.5 mL/kg. the mortaltiy of LPS-changed mice decreased 31.25%. Clinical symptoms and organ histological changes in HJM treated groups were decreased at a certain extent.3 Serum TNF-a, IL-6 levels and liver NO contents significantly depressed (P<0.05 or P<0.01), IL-4 and PGE2 level increased (P<0.05 or P<0.01) as compared to those of endotoxemia control group. Serum IL-1 level depressed, and liver SOD activity increased, but not statistically significant.ConclusionHJM remarkably decreased mortality of experimental mice, relieve clinical symptoms and organ histological changes, which may be related to its decreasing serum TNF-a, IL-6 levels and liver NO contents, and increasing serum IL-4 during endotoxemia. These findings provide a new strategy for the treatment of endotoxemia.
