The Effect Of The Combination Of Weifuchun And Retinoic Acid On The Treatment Of Gastric Precancerous Lesions And The Expression Of Rb Gene And C-erbB-2Gene

Objective: To investigate the effect of the combination of Weifuchun Pill andRetinoic Acid Tablet on precancerous lesions of gastric carcinoma and its regulativeeffect on the expression of the Rb and C-erbB-2gene.Methods: One hundred and twenty-two patients with endoscopically andpathologically confirmed mild,amoderate and severe dysplasia with or withoutintestinal metaplasia were divided randomly into the treatment group and controlgroup.The treatment group consisting of63patients was treated with WeifuchunPill，Retinoic Acid Tablet，Omeprazole and Sucralfate. The other59patients weretreated with Omeprazole and Sucralfate as control. After three months of treatment,pathological specimens obtained by endoscopic biopsy were made pathologicaldiagnosis according to the standard. Rb and C-erbB-2protein were detected byimmunohis-tochemical method before and after the treatment.The SPSS17.0statisticalsoftware was used for statistical analysis.Results: Medication for three months, gastric epithelial dysplasia wasimproved significantly in the treatment group by the pathological efficacyrate68.25%.Compared with the rate37.29%of the control group,there was significantdifference (P<0.05).As can be seen that the combination of Weifuchun and RetinoicAcid can reverse gastric epithelial dysplasia.The positive rate of Rb gene rised from74.60%to88.89%after three months treatment.The difference was statisticallysignificant.(P<0.05).The expression intensity of Rb gene was significantlyincreased(U=2.543,P<0.05).Contras-ted to the the treatment group the expression ofRb gene just has little change in the control group. The difference between thetreatment group and control group was statistically significant(P<0.05). It can be seenthat the combination of Weifuchun and Retinoic Acid could up-regulate the expression intensity of Rb gene.Contrarily,the expression rate of CerbB-2genedecreased from34.92%to12.70%and the expression intensity of CerbB-2gene wassignificantly decreased（U=-3.032,P<0.05) in the treatment group.There was littlechange in the expression rate and the expression intensity of CerbB-2gene in thecontrol group.As can be seen that the combination of Weifuchun and Retinoic Acidcould down-regulate the expression of gene Cerb-B-2.According to the results,thetendency could be seen that the expression of gene Rb was increasing and theexpression of gene C-erbB-2was decreasing with the improving of the gastricprecancerous lesions.The serious side-effects of the combination of Weifuchun andRetinoic Acid has not been found during treatment.Conclusion: The combination of Weifuchun Pill and Retinoic Acid Tablet waseffective in treating gastric dysplasia.It could reduce the risk of carcinogenesis ofgastric precancerous lesions by reversing the gastric mucosa lesions and regulating theexpression of tumor suppressor genes Rb and oncogene C-erbB-2.
