Background: The pathogenesis of lung cancer is very complex, cellular immunedysfunction may be one important reason for its incidence and closely related to thepatient’s prognosis. The cellular immunity is mainly mediated by T lymphocytes.CD28、CD152can promote the activation of T cells and play a significant role in immuneregulation. Whether abnormal expression of CD28、CD152in the peripheral blood ofpatients with advanced lung cancer induce “the T lymphocyte immune incompetence anddecreased immune function for further?Objective: I have detected the CD28/CD152expression in peripheral blood ofadvanced lung cancer patients and explored CD28/CD152immunity in the pathogenesisof lung cancer. All these can provide a theoretical basis for immunotherapy of lungcancer patients.Methods: I have collected the advanced lung cancer patients of Yan’an UniversityAffiliated Hospital in April2011—Jan201240cases and22healthy volunteers in thesame period of Yan’an University Affiliated Hospital Medical Center physicalexamination. they were divided into two groups, the lung cancer group and the healthycontrol group. I have detected the expression of CD4~+CD28~+、CD4~+CD152~+、CD8~+CD28~+、CD8~+CD152~+T cell subsets etc in their external weeks fasting morningblood by flow cytometry and studied all the datas using spss13.0software for statisticalanalysis.Results:1.The CD4~+T cell percentage of T cell subsets and CD4/CD8ratio with40advanced lung cancer patients were significantly lower than healthy control group (t=29.214, P=0.000; t=20.662,P=0.000),while CD8~+T cell percentage with advancedlung cancer patients was higher than the healthy control group, but the difference wasnot statistically significant(t=-1.637,P=0.107). 2. The CD4~+CD28~+、CD8~+CD28~+T cell subsets with advanced lung cancer patientswere significantly lower than the healthy control group in peripheral blood (t=14.086,P=0.000; t=6.021,P=0.000),but CD4~+CD152~+、CD8~+CD152~+T cell subsets withadvanced lung cancer patients were significantly higher than the control group(t=-3.065,P=0.003; t=3.221,P=0.002).3. The express of CD4~+T cell subsets in advanced non-small lung cancer group waslower than the small lung cancer group, but the difference was not statistically significant(t=-1.603,P=0.117);The express of CD8~+T cell subsets in advanced non-small lungcancer group was higher than the small lung cancer group, but the difference was notstatistically significant (t=1.145,P=0.268).Conclusion:1.The CD4~+T cell percentage and CD4/CD8ratio in peripheral bloodof advanced lung cancer patients were significantly lower than the healthy control group,suggesting that immune dysfunction was in advanced lung cancer patients.2. The CD8~+T cell percentage in peripheral blood of advanced lung cancer patientswas slightly higher than the healthy control group,no significant difference.3.CD4~+CD28~+、CD8~+CD28~+T cell subgroup of advanced lung cancer patients weresignificantly lower than the healthy control group, CD4~+CD152~+、CD8~+CD152~+T cellsubgroup of advanced lung cancer patients were significantly higher than the healthycontrol group. They indicate that abnormal expression of CD28/CD152which isantagonistic signaling molecules may be one of the reasons of significantly lowerimmune function with advanced lung cancer patients.4.The lung cancer is divided into non-small lung cancer group and small lung cancergroup according to the histopathology, CD4~+T cell and C D8+T cell showed nosignificant difference in the two groups, suggesting that the immune function was noobvious abnormalities in the two groups.
