Object: observing the effect of adipose tissue derived mesenchymal stem cellscombined with self-assembling peptide repair acute myocadial damage.Method:Rat ADSCs were isolated and cultured in vitro.Rat left anterior descendingbranch was ligated to establish acute myocardial infarction models. FollowingsuccessfuI modeI establishment,40SD male Rat were assigned to PBS group,ADSCs group,SAP group and ADSCs/SAP group(n=8)。In the ADSCs group,107ADSCs were suspended in the100ul of the PBS and the mixture were infused intothe surrounding sites of the infracted region by4points at30min after MI. In PBS andSAP group, the same region was injected with an equal volume of PBS and SAP,25ulper point.In the ADSCs/SAP group.100ul mixtual solution were injected into thesame region. At2week after transplantion, transthoracic echocardiography wasperformed for evaluating the recovery of cadiiac function. Soon afterwards, the heartwas obtained undergo paraffins sections and frozen-section examination for Massonstaining and immunofluorescence,which was used to examine the VWF antibody、a-SMA and vimentin antibody.Result and conclusion: The primary ADSCs cells were obtained by zuk’smethods.Their morphology under light microscopy show similarly spindle-like shapeand gradually change with the addition of generation. Flow cytometry detection thatcells express CD29and CD44surface markers and vague expression of CD106andCD105. Immunofluorescence and Oil Red O staining confirmed that ADSCs can bedifferentiated into a-SMA,vWF,vimentin and Fat. DiI dye was uesd to track ADSCswhich were transplanted into ischemic myocardiumafter MI30min.The retention ratewas significantly higher in the ADSCs/SAP group than that of the ADSCs group at2weeks.At2weeks after transplantion, echocardiogram showed that ejection fraction(EF) and fractional shortening (FS) in treatment group (ADSCs, SAP and ADSCs/SAP) have a significant improvement than PBS group(37.3±8.3%and15.7%±4.1%;.Moreover, the EF and FS was higher in ADSCs/SAP group(71.2±6.2%and36.0±4.6%) than in ADSCs group(49.9±9.7%and22.38±5.30%) andSAP(47.6±9.9%and23.2±6.70%) group. Masson’s trichrome staining displayed thatthe scar area were lower in ADSCs and SAP group than in PBS group.therefore, inADSCs/SAP group is higher thanin ADSCs group or SAP group.Immunofluorescence revealed significant improvement in the vessel density inADSCs or SAP or ADSCs/SAP group compared with PBS group(P<0.05~0.01)。From the above, adipose tissue derived mesenchymal stem cells combined withself-assembling peptide therapy obviously repair not only injured myocardium,butalso can reduce fibrinous exudate and promote neovascularization.
