Effect Of OK-432and PolyI:C On Maturation Of Mouse Dendritic Cells And Against Injury

Objective To investigate the effect of OK-432and Poly (I:C) on the maturation ofmurine bone marrow-derived dendritic cells and co-culture with supernatants ofMouse forestomach carcinoma(MFC).To study the protective effect of OK-432in acombination with PolyI:C on DCs against injury and to speculate the possiblemechanism of this function.Methods The immature DCs were generated from mouse bone marrow monocytesin the presence of recombinant mouse GM-CSF and recombinant mouse IL-4.TheseDCs were then pulsed separately with OK-432and Poly(I:C) or in combination. DCswere co-cultured with different concentrations of MFC supernatants for24hours.Morphological characters of DCs were observed under the electron microscope. DCssurface molecules CD83and CD86were assayed by flow cytometry.The IL-12levelsin supernatant were assayed by ELISA. The appearance of dendritic cells afterco-cultured with MFC supernatants for24hours under optical microscope. Antigenpresenting ability of DCs in allo-MLR and survival rate of DCs were measured byMTT assay. Zinc finger protein A20(A20) expression in DCs was determined byimmunohistochemical procedures.Results The DCs pulsed with OK-432and Poly(I:C) showed typicallymorphological characters, expressed much more antigens CD83and CD86on thesurface, significantly higher secretion of IL-12and proliferation of lymphocytes thanDCs with other treatments; The supernatants of MFC could induced the damage ofDCs. Increased level of survival rate and A20protein in the union group of DCs werefound.Conclusion OK-432in a combination with Poly(I:C) can effectively inducematuration DCs；OK-432in a combination with Poly(I:C) can enhance the survivalrate of DCs，which is associated with the increased expression of A20protein.
