Objective To investigate the feasibility of establishing an animal model of intervertebral disc degeneration induced by needle puncture in the anulus fibrosus; And to observe the effect and the regularity of the cytokines in the Rabbit Degenerated Intervertebral Disc With Transplantation of Bone Mesenchymal Stem Cells;Methods (1) A total of32New Zealand white rab were selected,24of them were operated and exposed to the lumbar intervertebral disc of L3/4> L4/5and L5/6, the discs were injured by16-gauge needle at a depth of about5mm to puncture the right anterolateral anlllus fibrosus。After surgery, by random assignment, the24rabbits were divided into3groups:blank group (B group)、interventions group (C group) and experiment group(D group), The rest8rabbits were just exposed to the lumbar intervertebral disc, but no puncture, and as the normal group (A group); The imageological change of discs, which were selected2ones from every group by random assignment, was studied by MRI at4weeks after the operation, and to observe the degeneration of the lumbar intervertebral discs;(2) Tranplanted the BMSCs which were developed to P3generation into the degerated discs of D group, the same amount of alginate were transplanted into C group, while A and B groups without any handling;(3)At2、4、8、12weeks, respectively selecting2rabbits fron every group, using RT-PCR to measure the expression value of MMP-1、MMP-2、 MMP-3、TNF-α、IL-1and TGF-β, while Weston Blot for the Proteoglycan and the II Collagen;(4) SPSSI2.0analysis the datas。Result (1) All the discs were degeneration by MRI;(2) The expression value of MMP-1、MMP-2、MMP-3、TNF-α and IL-1were higher in B、C group than nomal discs of C,TGF-β、Proteoglycan and the II Collagen were lower(P<0.05); and after tranplantion the former five cytokines were lower than the degenerated intervertebral disc (P<0.05), with the time of tranplantion, the tendencies more and more obviously (P<0.05), but the TGF-β、Proteoglycan and the Ⅱ Collagen was adverse (P<0.05)。Conclusion (1) The anulus needle puncture can induce the progressive degeneration of the intervertebral disc in rabbits, this model is available for the clinical study of degenerative diseases of the intervertebral discs;(2)The expression value of MMP-1、 MMP-2、MMP-3、TNF-α and IL-1were higher in degenerated intervertebral discs than nomal discs, TGF-β、Proteoglycan and the Ⅱ Collagen were lower;(3) After tranplantion, MMP-1、MMP-2、MMP-3、TNF-α and IL-1were lower than the degenerated intervertebral disc and with the time of tranplantion, the tendencies more and more obviously, but TGF-β Proteoglycan and Ⅱ Collagen were adverse。...
