Expression And Significance Of15-LOX-1in Colorectal Cancer

OBJECTIVE： Studies indicated that15-LOX-1plays an important rolein the development of tumors. But the role15-LOX-1playing in colorectalcancer is still controversial. This study was designed to evaluate theexpression of15-LOX-1in colorectal cancer, and the correlation of itsexpression to clinical characteristics and prognosis of patients withcolorectal cancer，and search for the possible mechanism.METHOD: By adopting immunohistochemistry, the study investigatedinto the expression of15-LOX-1in103cases of colorectal cancer tissueswithout the history of radiotherapy or chemotherapy，and56cases of normalcolorectal mucosa adjacent to tumor nests．The results were then analyzedin consideration of the clinicpathologic features, the prognosis ofpatients and others indicators regarding invasion and metastasis.RESULTS:（1）Expression of15-LOX-1was56.3%in colorectal cancertissues，considerably lower than that in normal colorectal mucosa adjacentto tumor nests，which was73.2%（P＜0.05）．（2）The expression of15-LOX-1was closely related to histological grade，Dukes’stage，lymph nodemetastasis，and distant metastasis (P＜0．05)．Low expression of15-LOX-1was detected at the advanced stage of cancer or on colorectal cancerpatients with lymph node metastasis，and distant metastasis（3）The1-,3-and5-year survival rates of15-LOX-1-positive patients with colorectalcancer were significantly higher than that in patients with negativeexpression(P＜0．05) and these15-LOX-1-positive patients appeared tolive longer．Cox multivariate analysis showed that15-LOX-1and lymph nodemetastasis can be used as independent prognostic indicators(P＜0．05)．（4） The expressions of15-LOX-1was inversely correlated with the expressionsof VEGF、MMP-2、MMP-7in colorectal cancer tissues (r=－0.360、－0.374、－0.346，P＜0．05)．CONCLUSION:（1）15-LOX-1played a positive role in containingcolorectal cancer, and it may be an important indicator in biologicbehavior and prognosis prediction of colorectal cancer.（2）The antitumoreffect of15-LOX-1playing in colorectal cancer may be associated withVEGF and MMPs.
