ObjectiveTo study antifungal effects of the Two-component Signal TransductionSystem inhibitors ethacridine and closantel against aspergillus,fusarium and Candida albicans in vitro, so as to prepare for thedevelopment of a new efficient and safe antifungal.Methods1. The minimal inhibitory concentration (MIC) of ethacridine,closantelor amphotericin B alone was determined against aspergillus or fusariumby NCCLSM38-A liquid-based microdilution method; The minimalinhibitory concentration (MIC) of amphotericin B was determinedrespectively when amphotericin B combine with ethacridine or closantelagainst aspergillus or fusarium in the same way.2. Estimate the growth concentration of Candida albicans in RPMI1640liquid culture at different times with the effect of ethacridine andClosantel, and draw time-antibacterial curve.3. Detect the rate of Candida albicans’adhesion to oral epithelial cellunder the action of Two-component inhibitors ethacridine and closantelwith different concentrations.Results1. Ethacridine and closantel have the characteristic of inhibiting thegrowth of aspergillus and fusarium in vitro. Aspergillus and fusariumare hypersensitive to amphotericin B with the help of ethacridine orclosantel.2. Ethacridine and closantel can inhibit the growth of Candida albicansin different degrees, and the inhibition enhanced as the concentrationincreased; but the same concentration between ethacridine and closantel have no statistical significance.3. Ethacridine and closantel with different concentrations caneffectively deduce Candida albicans’adhesion to oral epithelial cell,the influence to adhesion enhanced as the drug concentration increased.The same concentration between the two drugs have statisticalsignificance, ethacridine have more significant impact to the effectof adhesion.ConclusionsTwo-component Signal Transduction System is a new antifungal drugtarget; Ethacridine and closantel have antifungal properties againstfungi in vitro and they are promising anti-fungal drugs.
