The elasticity of cell is an important parameter of cell mechanics, cell structure and function are closely related to the mechanical properties of cells. Outside the physical environment changes, such as the substratum hardness that the cell was cultured change will lead to change of the elasticity of cell. The atomic force microscopy(AFM) has been an extremely important tool for cell mechanics due to its ability to measure sample properties in physiological conditions with minimal destruction.In this thesis it focused on preparation of different hardness of substratum films with chitosan, the study of the influence about the chitosan films and the cell by AFM. Through the chitosan solutions with different concentrations, the use of cast preparation of chitosan films of experiment required, and to evaluate the cell compatibility, the swelling index, contact angel, hardness of chitosan films. Normal endothelial cell and Hepatocellular Carcinoma Cell were seeded on different chitosan films to test cell morphological characteristics(like cell spreading area, cell length and minor axis ration) and the elasticity of cell. The following are the results of the present study:①The findings show that the swelling index of chitosan films is the largest is3%and the least is4%; the contact angle of chitosan films are about65°~81°, the hardness of chitosan films are about315KPa~413KPa, they are both increased with the increase of the concentration of chitosan solution that films are prepared; The experiments that cell compatibility of chitosan films shows that chitosan films have good compatibility;③the cell spreading area of endothelial cell and Hepatocellular Carcinoma Cell are measured, it increased with the increase of the concentration of chitosan solution that films are prepared; the ration of length axis and minor axis of cells are different in different time, it shows that the cell morphology are changed on different chitosan films; the largest elastic modulus of endothelial cells is about2.44KPa and the least is about1.75KPa, the largest elastic modulus is about1.4times the least; the largest elastic modulus of Hepatocellular Carcinoma Cell is about1.32KPa and the least is about0.66KPa, the largest elastic modulus is about2times the least; and the elasticity of the cells both are increased the increase of the concentration of chitosan films are prepared. The findings illustrates that different chitosan films can affect the elasticity of the cells has clearly influenced. The findings will contribute to the explanation for the mechanism of the interaction between chitosan and cell, and provide the theoretical support for the further study and application in Cell Biology.
