In traditional Chinese herbal medicine, Lycopodium obscurum L. has been usedfor treatments of bone fracture, traumatic injury and contusion. Its efficacy has beenconfirmed by modern clinical medicine. However, the effective components is unclear.Recently, a new compound Serratenediol has been separated from Lycopodiumobscurum L.. This study is to explore the effect of Serratenediol on the osteogenicactivity of cultured osteoblasts in vitro.In this study, the effect of Serratenediol on the osteoblast proliferation, ALPactivity, the expression of osteogenic gene in osteoblast was studied.MTT assay result revealed that Serratenediol could enhance the osteoblastproliferation rate in early treatment.The result showed that that Serratenediol could enhance the ALP activity ofosteoblast at the concentration of2μmol/L、4μmol/L in the treatment of9daysReal-time PCR result showed that: in early treatment period(3days)Serratenediol could stimulate the expression of c-Fos、c-Jun、Runx2, but inhibited theexpression of Fra-1、Fra-2and Osterix.At the6days treatment it could enhance theexpression of Fra-1、Fra-2and Osterix. Long treatment period (9days) could enhancethe expression of Osterix, ALP, BSP, OPN and Col-I but inhibited the expression ofFra-1, Fra-2and Runx2.The above result demonstrated that Serratenediol could modulate osteoblastproliferation rate, ALP activity and the expression of osteogenic gene in osteoblast.Serratenediol may be one of the effective compositions of Lycopodium obscurum L.for the treatment of bone disease.
