Objective: To investigate the effect of cisplatin on the expression of Matriptase,uPA-mRNA in human endometrial cancer RL-952cells.Methods: Culture human endometrial cancer RL-952cells in vitro, and detectthe effect of cisplatin on the expression of Matriptase, uPA-mRNA by reversetranscription polymerase chain reaction (RT-PCR).Results:The studies showed positive expression of Matriptase, uPA-mRNA in humanendometrial cancer RL-952cell lines.Cisplatin could cause the significantly decreaseof the expressions of Matriptase, uPA-mRNAs in dose-dependent manner after24hours incubation in comparison with the control group(P<0.05).After incubationwith8mg/L cisplatin in different time-points, the expression of Matriptase-mRNAdecreased obviously in comparison with the control group after4hours (P<0.05),when the expression of uPA mRNA decreased obviously after8hours(P<0.05), therewas obvious time-dependent manner between them.Conclusions:The studies showed positive expression of Matriptase, uPA-mRNA in humanendometrial carcinoma RL-952cell lines.Cisplatin could inhibit the expression ofMatriptase, uPA-mRNA of human endometrial carcinoma RL-952cell lines in aconcentration dependence. After incubation with8mg/L cisplatin, the down-regulationof Matriptase-mRNA occurred earlier than uPA-mRNA.
