The immune system is important to implement the immune responses and plays the immune functions, including the immune organs, cells, and molecules. The immune system can protect self, and reject non-self. Once pathogens invade the body, they can be presented to T cells by the antigen presenting cells (APCs). Then the cellular and humoral immunity can be active, and removes the invading pathogens outside. Therefore, as the central link in the immune response, APCs in the process of self-protection function of immune system play an important role. Meanwhile, the function of APC regulation, especially the down-regulation of APC plays important roles in maintaining immune homeostasis.Macrophage (MΦ) is one of the important cells in the innate immunity, its main functions include phagocytosis, antigen presentation, and secreting some cytokines. MΦcan devour and eliminate the invading bacteria, viruses and other infectious microorganisms, which can be presented to antigen-specific T cells and activated T cells by pMHC-TCR and costimulatory molecules. In this process, MΦalso secretes IL-1, IL-12 and other cytokines, which constitute the necessary micro-environment to eliminate external antigens.MΦmakes the key role in the immune response of various immune diseases. It becomes the focus of the researches. Some studies suggest that excessive activation of MΦis an important reason leading to rheumatoid arthritis. In China, traditional medicine used in the treatment of autoimmune diseases have been about 2000 years, especially rattan plant has been widely used in folk. However, the mechanism is not clear at present. So, we extracted one compound from the wild ratten plants, named as T2009.In our studies, we observed a variety of immune cells in mice by T2009. We found only MΦvacuolization obviously, not other immune cells. In vivo, the function of phagocytosis decreased when the mice were injected T2009.Ia,CD40,CD80,CD86,CD11b and other molecules related to antigen expression levels were lower than the mice not injected drug. Meanwhile, the activation and amplification of CD4+T cells also reduced, and drug T2009 inhibit gene transcription of murine peritoneal macrophages IL-1, IL-12p35. On the basement of some vitro experiments, the studies in vivo demonstrated the best evidences that drug T2009 could down-regulate the immune responses.In these experiments, we revealed the MΦdown-regulations after the Drug T2009, using mice models tests. We believe that T2009 on the treatment of autoimmune diseases, as the cell specific drug, have widely prospect. It is worth studying furthermore.
