The ecological environment of some karst areas in southwest of China is very difficult, the soil erosion is serious, the contradiction between human and land is prominent, introducing lonicera japonica has become a kind of important method of vegetation restoration in these regions. The Mengjin1st in Chongqing is introduced in Shandong; this kind of lonicera japonica has played an important role of ecological restoration in karst rocky desertification areas of Chongqing. This paper has studied the medicinal ingredients of the Mengjin1st in its country origin Shandong and4karst rocky desertification regions of Chongqing. In addition, it has compared the determination results of the two methods and contrasted the differences of two methods; besides, preliminary analyzed the main problems of the UV method and put forward some ways to improve the accuracy of the UV detection, and then outlook the application prospect of the UV and HPLC method in lonicera japonica’s quality inspection.The results of this paper:(1) In the result of chlorogenic acid content determined by UV method:the highest content is the lonicera japonica sample in Xiushan(N0.3), the lowest is in Shandong(NO.1); in the result of luteoloside content:the highest is in Xiushan(N0.3), the lowest is in Chengkou(NO.6) and Shandong(NO.1).In the result of chlorogenic acid content determined by HPLC method:the highest content is the lonicera japonica sample in Xiushan(NO.3), the lowest is in Shandong(NO.1); in the result of luteoloside content:the highest is in Nanchuan(NO.5), the lowest is in Shandong(NO.1).(2)This paper has used two methods to determine the chlorogenic acid and luteoloside content in lonicera japonica, according to the comparative analysis of the determination results of them, The content of chlorogenic acid and luteoloside determined by UV are both higher than HPLC method, the chlorogenic acid contents determined by UV are mostly2-9times than HPLC, the luteoloside contents determined by UV are mostly24-27times than HPLC.(3) Analysis the reasons that the results determined by UV method is different from the HPLC method, point out that the main problems of the UV method is that this method is without separation, so the result determined by UV is the total content of the same chromophore in lonicera japonica, but not a single component content, on the contrary, the measured result of HPLC method is the content of a single component, because the content of the medical components are determined after the components of the lonicera japonica are separated in the chromatographic column, so the content results of UV are higher than that of HPLC.(4) It points out that the accuracy of the results determined by UV method can be improved by optimizing extraction method and using some separation technology, because of the chromatographic interference of the coexistence, its lack of the key process of separation makes UV can’t determine the contents of chlorogenic acid and luteoloside accurately alone, so using some separation technologies is an effective method to improve the UV method’s detecting accuracy.(5) Obviously, HPLC method can effectively separate the chlorogenic acid and luteoloside from their analogues and eliminate the interference, so the application prospect of HPLC are more vast than UV in detecting medicinal component content of lonicera japonica. As a common method in laboratory, the UV method, in order to get an extensive use, can be used together with some separation technologies and other new technologies in future.
