Nanotechnology, as a new filed, become the three major research area of the word withthe information technology and biotechnolodgy. In this research, enrofloxacin nanoemulsionwas prepared using palmitic acids and enrofloxacin, which nanoparticle was111.07.2nm,polydispersity index was0.26±0.17, electromotive force was-31.573.76mv and the drugof enrofloxacin was5.0mg/mL. The experiment of feeding pigs and the real-time PCR injejunal epithelial cells (IPEC-1) are investigated, the main contents are as follows:1. The experiments of the growing pigs in vivo, enrofloxacin nanoemulsion taked to the pigsaccording to0.6,1.25,2.5and1.25mg/kg respectively, determinating the blood biochemicalindex after6,24,48h, the antioxidant index and the histopathological observation. The resultsshowed that the content of MDA, biochemical index and the antioxidant index had nosignificant effects on the liver, kidney and pancreas by oral enrofloxacin nanoemulsion(P>0.05). Any how it had no influence on the organs of the pigs.2. The experiment of IPEC-1cell activity, using MTT method determined the cell viability,showed that the cells had no effect with the dose of5μg/mL enrofloxacin nanoemulsion, butthe cell inhibition was enhanced with the increasing dose.3. The experiment of Escherichia coli inhibition, the enrofloxacin nanoemulsion wasinvestigated on the E. coli. The results showed that the MIC is0.078mg/mL, susceptibilitytesting showed that inhibitory significance from enrofloxacin nanoemulsion was the same asoriginal drugs, still keeping inhibitory availablity of the enrofloxacin.4. The experiment of real-time PCR, using the concentration of1,10,50and100μg/mLrespectively in the IPEC-1cells, collected the culturing cells at6,12,24and48h, andinvestigated the mRNA expression of IL-6and TNF-α. The mRNA expression of IL-6andTNF-α were increased with compared to the control group, while the IPEC-1cells with10μg/mL doses after24h. The cells of50μg/mL and100μg/mL became apoptosis and themRNA expression of IL-6and TNF-α were gradually reduced.This research will provide a apply model in vitro for nanomaterious drugs in intestinal tract.There are value on Nanobiotechnology and nanomedicine, nanocomposites and applications,modeling and simulation of nanostructures.
