In this experiment, Wistar rats were used as experimental animals, we investigated low lever laser therapyacts on adhesion molecules in a model of mastitis induced by lipopolysaccharide in rat. The purpose is toreveal the migration patterns of PMN. In order to prevent mastitis on the clinical application of low lever laser,and lays an experimental basis.First, setting up the test rat mastitis model, then injection LPS induced mastitis from the fourth pair ofmammary.180pregnant Wistar rats were randomly divided into four groups. Low lever laser therapy in groupA after LPS breast perfusion; Just breast perfusion of the same amount of LPS in group B; low lever lasertherapy in group C after injection of stroke-physiological saline solution on the fourth pair of mammary; onlythe same volume of stroke-physiological saline solution in group D. Group A and C rats laser irradiated afterperfusion in1h,15min each time, twice each day, seven days of continuous irradiation. Group B and D was notirradiated, anesthetized the rats quickly Respectively in perfusion after injected LPS in6h,12h,24h,48hand7d and clipped the fourth breast, then abdominal aortic blood in collection tubes, some addedanticoagulants for separating PMN, the others without anticoagulants for the separation of serum, and finallythe rats were put to death. Flow cytometry detection of CD11b, CD62L by separation form the PMN, ELISAmethod to the IL-8, IL-1beta and TNF-alpha, those were cytokines, detected by separation from the serum. Thetotal RNA was extracted from the breast tissue by using Fluorescence quantitative PCR to detect the mRNAexpression level of ICAM-1and PECAM-1.Test results:(1) It can induce ideal mammary inflammation reaction with the40μg LPS in each side of themammary grand, so the mastitis model successfully copied, can be used in subsequent experiments.(2) lowlever laser irradiation can reduce the content of blood level ofIL-1β and IL-8after injection LPS24hours.(3)After low lever laser irradiation, the content of CD62L was inhibited to reduce after injection LPS24hoursand48hours.(4)The low power laser irradiation has made the mRNA expression of ICAM-1decreased afterinjection LPS12hours、24hours and48hours.Conclusion: After low lever laser irradiation, it can reduce inflammatory cytokine IL-1beta, the release ofIL-8and inhibition of CD62L, and it can inhibit the mRNA expression of ICAM-1at the same time, thus it canreduce the PMN adhere to platelet-endothelial cell, in the result of reducing the mammary gland tissuedamage.
