The GP3protein is one of the minor structural proteins expressed by porcine reproductive andrespiratory syndrome virus (PRRSV). GP3is a heavily glycosylated glycoprotein which is encodedby the viral open reading frame (ORF)3. Although not much information on the role of the GP3protein during infection of cells with PRRSV is available, more and more evidence indicates thatits main function is related to viral replication, assembly, and contribution to viralheterogenicity.For that reason,we first prokaryotic expressed GP3protein which is coded by ORF3PRRSV,and prepared polyclonal antiserum from it; at mean while, we used phage12-mer peptide libraryto screen GST-GP3protein. At the end we have one dodecapeptide, the sequence isSVSVGMKPSPRP, we named it S polypeptide, and synthesized it. The ELISA result suggestedthat S polypeptide can inhibite PRRSV virus from infecting cell, and the ratio of inhibition risedwith the input of S polypeptide.In conclusion, this research offered reference for identifying PRRSV from other porcine diseaseand GP3functional receptor, and provide basic data for PRRSV prevention and small moleculesbiologics.
