Lycopene is one of the tomato processing quality traits.The phenotype of lycopene wascontrolled by more than one factor pairs, and the genetic mode of lycopene fits the quantitativetrait locus, such as Del、hp、Ogc、dg、B、t and so on. hp and dg genes can significantly improvethe tomato lycopene content.To improve the level of lycopene content, it is an effective way tobreed a cultivar carrying more than one pair of genes. With the molecular polymerizationdevelopment of breeding technology, marker-assisted selection (MAS) technology can aggregatemore than two excellent traits, which improve breeding efficiency.MAS is an efficient method toselect plant carrying target gene compared with traditional breeding work.This study identified the SNP molecular marker linked to hp-1、hp-2and dg genes,and established the Multiplex-PCR reaction system, which has been applied in selection ofhomozygosis genotype of F2plants. Combinations of hp-1and hp-2and those of hp-1and dg genes were made in the same homozygous plants.1.The specific primers were designed according to the nucleotide mutation in hp-1、hp-2anddg sequences of NCBI, and the target fragments were amplified by the reaction system65designed.The result was stable and reliable.2.Double-PCR reaction systems for hp-1/hp-2and hp-1/dg genes were established. In thehp-1\hp-2reaction system, the TM temperature was54℃, the primerconcentration ratio was1.8:1.0; In the hp-1\dg reaction system, the TM temperature was51℃, the primerconcentrationratio was2.0:1.0.3.Two homozygous plants carrying hp-1and hp-2genes were selected from307F2plants; and3homozygous plants carrying hp-1and dg genes were selected from342F2plants.4.The results of determination of lycopene content and other physiological indexdetermination showed that, the content of lycopene in homozygous plants with hp-1/hp-2andhp-1/dg, was increased by15.3%and20.0%than their parents respectively;36.3%and32.0%thanCK plants respectively. Other physiological index showed little changes in the experiment.
