| Pelodiscus sinensis, as a kind of aquatic product with rich nutrition and medical value is favored by modern increasingly. With the deepening of the concept of a healthy diet, people in the pursuit of the nutritional value of aquatic products also now began to focus on the safety of aquatic products, a more healthy culture mode becomes the inevitable development direction of Pelodiscus sinensis breeding.The trial was mainly based on healthy breeding base in Chongming, Pelodiscus sinensis breeded around paddy so as to realize paddy with Pelodiscus sinensis survival together.Compared with normal paddy field and fish pond, the pond water and sediment microbial diversity analysised mainly by means of PCR-DGGE technology, and in combination with microbial count results and physical and chemical character such as DO,COD and so on for comprehensive evaluation,in order to provide experimental basis for healthy, stable, efficient aquaculture mode.In addition to provide reference to paddy together with Pelodiscus sinensis,sediments of different culture pond in Shanghai jinshan aquaculture base had been researched.The physical and chemical data of Chongming showed that Pelodiscus sinensis around paddy pond rendered on a similar change rule on COD,TP,DP,TN and NH4+,it indicated that there is a certain correlation existed between different index. And fish pond had good stability on conductivity,DO,TP,DP,PP, NH4+and NO2-,and this indicated water quality was more stable in fish pond.Microbial count analysis of aquatic water in Chongming showed that from August to November the quantity of mould and yeast in Pelodiscus sinensis around paddy pond maintained at the level in103CFU/mL, and the quantity of lactic acid bacteria was basically stable at103CFU/mL.The quantity of various bacteria in paddy field was maintained at a high level and stable,and fish pond at a low stable level.Similarity analysis of microorganism species by PCR-DGGE showed that the similarity of bacterial species between two sampling points of Pelodiscus sinensis around paddy pond was higher, which reached the highest similarity to92.9%in November,fish pond was different from other pond and the similarity was only42.4%in November.The closest species classification from sequence alignment demonstated that pedobacter glucosidilyticus, flavobacterium, proteobacterium, bacteroidetes bacterium,chryseobacterium and actinobacterium had been detected,Some of flavobacterium and chryseobacterium were conditioned pathogen,so needed to be monitoring in the process of breeding.Microbial count analysis of aquatic sediments in Chongming showed that the quantity of bacteria,yeast,lactic acid bacteria and organic phosohate-solubilizing bacteria increased to105CFU/g in october,and the bacteria quantity in paddy field and fish pond was same with aquatic water. Similarity analysis of microorganism species showed that the similarity of bacterial species between two sampling points of pelodiscus sinensis around paddy pond was higher, which reached the highest similarity to91.4%in August, fish pond was different from other pond and the similarity was only13.1%in November. The closest species classification from sequence alignment indicated that flavobacteriaceae, sphingobacteriaceae and pedobacter terricola existed in these sediments.Microbial count analysis of aquatic sediments of Penaeus vannamei in Jinshan shanghai aquaculture base showed that the number of bacteria,yeast,mould and lactic acid bacteria maintained stable in bog and drain from June to October,it indicated that bog had great advantages in maintaining the stability of the ecological environment. The number of bacteria and yeast in grass grouded pond was stable in each months.The number of lactic acid bacteria and actinomycetes stayed stable in oxygen added tiny bubbles pond. The DGGE technology combined with sequencing results showed that there were few advantage bacterium group in the first two months,but increased in the later months,this maybe because of excessive feeding,and it caused the deposit of organism, remaining bait,excreta and other toxic and harmful substance, and created a conducive proliferation environment to bacterial.Therefore, intense prevention needed to pretect Penaeus vannamei in the later period so as to avoid unnecessary loss. |
