Flax is an important economic crops widely used. plant height and processlength is positively correlated with agronomic traits and fiber yield, but also theimportant breeding objective. Brassinosteroid (BR) are plant steroid hormones thatregulate a variety of plant growth and developmental processes, especially play aimportant role in cell elongation. BRI1is a crucial component of a transmembrane BRreceptor, and BES1is a key factor involved in the induction of BR gene expression. Inthis experiment, make use of Xinjiang main cultivated Flax Variety ’Fanny’ asexperimental materials, cloning the core fragment of LuBRI1and LuBES1gene of BRsignal pathway, and establishment of regeneration system of stable ’Fanny’organogenesis, for study the BRs on the role of regulation the plant height of flax,which lay the foundation for the improvement of fiber quality and yield of flax.According to the experimental results,(1) the CTAB method is more suitablefor flax DNA extraction, high quality DNA can be used to amplify the long fragment.Homologous alignment, and design of degeneracy primers, which was obtainedLuBES1and LuBRI1core sequence are832bp and1000bp by PCR respectively.Further obtained the core fragment of LuBES1by Site-finding PCR.(2) In20different of hormone combinations callus and shoot induction medium. In this study,the optimum medium for inducing adventitious buds was1/2MS+0.5mg/L6-BA+0.9mg/L KT+0.2mg/L IAA, induction callus rate reached99.7±0.65%,browning rate reached0.83±1.44%, budding number reached93.2±5.89%. Theoptimum medium for root inducing was1/2MS+0.3mg/L NAA+0.05mg/L IBA. Kanand Hyg for ‘FANY’ screening threshold was120mg/L and40mg/L.(3) Through thestudy on flax physiological experiments determination of GA3, ABA regulation offlax germination suitable dose of1mg/L and50mg/L, respectively. epiBL(2,4-epi-brassinolide) regulation of flax hypocotyl most appropriate for1000nM inthe dark. In the light, under the1nM epiBL hypocotyl is longes. Root under the optimum epiBL for100nM and1nM in light and dark. The above results lay thefoundation for later study of plant hormones to regulate growth and development toflax.
