Tomato is one of the most commonly edible fruits vegetables in people’s life, and carotenoid is very rich in ripe tomato. The research on tomato resource would have an important economic value for the rational development and utilization of agricultural and sideline products.In this paper, tomato was material for the research, and carotenoid in tomato was studied, the main contents included:(1) The extracting and determining technics for carotenoid from tomato were studied. Extracting and ultrasonic method were used to extract carotenoid in tomatoes, respectively, and the content of carotenoid was determined by spectrophotometric method. The single factor test and orthogonal experimental design were carried out to optimize the extracting technology for carotenoid in tomato. The result indicated that the effect of ultrasonic extraction was much better than the other by conducting a comparative study of the optimum condition for them.(2) The separation methods for carotenoid in tomato were studied. The extracting solution was separated by paper chromatography and column chromatography, respectively. The result indicated that both types of chromatography could not effectively separate the components of carotenoid.(3) The method for determining carotenoid in tomato was developed by high performance capillary electrophoresis (short as HPCE). The optimal experimental condition was as follows:detection wave length was450nm, running buffer was pH4.5, HAc-NaAc was20mmol/L, electrophoresis voltage was24kV, injection time was6s with the pressure of0.5MPa, electrophoresis temperature was20℃.The samples could be completely separated in14min with good linear relationship in the optimum condition.(4) The carotenoid of tomato in the red stage was determined under the optimal condition of HPCE, and it indicated that the lycopene and β-carotene contents were21.18μg/g and5.14μg/g,respectively. The feasibility of analytical method was good according to the results of precision, reproducibility, and recovery experiments.(5) The content of effective component in lycopene or β-carotene soft capsule was determined by HPCE. The menstruated content of lycopene was matched with the marked amount of lycopene soft capsule, and the menstruated content of β-carotene was similar to the marked amount of natural β-carotene soft capsule, therefore, it demonstrated to be an accurate method that was conducive to standardizing the carotenoid products market.(6) The samples of tomato in five different ripening stages were measured and analyzed in accordance with optimal condition of HPCE. The result indicated that the content of β-carotene was decreasing firstly and then gradually increasing with the content of lycopene was increasing continuously during the different mature phases, and both of them would get to the highest content until the fruit ripened completely.
