Avian influenza belongs to the orthomyxovirus virus family influenza virus, is apetent infectious poultry, caused by influenza A virus which can lead to human andanimal comorbid. The fact of that the incidence of death with highly pathogenic avianinfluenza virus infection human appears, human are increasingly aware of hazards ofAIV. In order to improve the disease resistance of birds fundamentally, people startedto pay attention to improve poultry disease resistance by means of breeding fordisease resistance.DNA methylation is a very important one of epigenetic modifications. Detectionof DNA methylation can assess gene expression to a certain extent, combined withpathogens infect the host when the host cell gene expression differences in levels ofanalysis, a preliminary understanding of the course of the process of change the keyeffects of genes and their regulatory mechanisms, can provide a reference for thesubsequent use of such genes in breeding for disease resistance.To reveal the effect of the infection of avian influenza virus on the genome-wideDNA methylation level of the host, the DNA methylation levels and patterns ofCCGG sites in genomes was analyzed among nine different tissues of SPF chicken,SPF chicken infected by A/Goose/Guangdong/1/96, H5N1and A/Duck/Anhui/1/2006,H5N1respectively, SPF duck, and SPF duck infected by A/Goose/Guangdong/1/96,H5N1and A/Duck/Anhui/1/2006, H5N1respectively. The main results are as follow:1. By replacing radiation with fluorescent system in the technique ofmethylation sensitive amplified polymorphism (MSAP) and optimizing reactionconditions, a modified technique to detect DNA methylation called F-MSAP(fluorescent labeled methylation sensitive amplified polymorphism) was developed..2. The total DNA methylation degree of the SPF chicken infectedA/Goose/Guangdong/1/96, is higher than the corresponding organization of thecontrol group of SPF chicken in heart, trachea, upper and lower trachea. While the total methylation level of SPF chicken infected by A/Duck/Anhui/1/2006of a chickenis lower than the control group, the corresponding organization in heart, spleen,trachea segment, the lower trachea, thymus and bursa.3. In the spleen, the methylation ratio difference trend of infected andnon-infected chickens, infected and non-infected ducks is the same, which theinfected chicken/duck is higher than the non-infected chicken/duck.However, thistrend is different in the thymus and bursa, which the DNA methylation level ofinfected chicken is higher than the control chicken, while the level of infected duck islower than the control duck.These results demonstrated that there was difference in the DNA methylationlevel between the chicken/duck infected by AIV and SPF chicken/duck, whichsuggested that the infection of avian influenza virus (H5N1) may have effect on theDNA methylation level of healthy hosts.
