Dry seeds of wheat Guo9722and Heke2hao were radiated by60Co γ-ray, Variations of protein content, gluten content, zeleny and hardness of M4and M5population were studied, screening variation lines. in M5mutant lines,Use Brabender farinograph to measure farinogram parameters, application to SDS-PAGE analyzed high molecular weight glutenins (HMW-GS),In order to investigate the radiation treatment effect quality characteristics of wheat, for wheat quality improvement to explore new ways.1Variations of protein content, gluten content, zeleny and hardness of Guo9722of341lines and Heke2hao of234lines in M4population were analyzed, According to the variation strains protein content and wet gluten content,based on a criterion out of the range of mean±2s,In Guo9722M4population, selected8with higher protein and gluten contents and the4with the lower r protein and gluten contents,There are5lines reach to the national high-gluten wheat standards.In Heke2hao M4population selected6with higher protein and gluten contents and the4with the lower r protein and gluten contents,6lines reach to the national high-gluten wheat standards.2The generation correlation analysis between M4selected strains and M5from Guo9722and Heke2hao showed that all four quality traits were positively correlated. generation correlation of protein content, wet gluten content and sedimentation value was up to0.01level in Guo9722, generation correlation of hardness up to0.05level; generation correlation of protein content was up to0.05level in Heke2hao. Two groups which contained high protein content and wet gluten content and low protein content and wet gluten content from M4still remained the same in M5, the trend of strains grouping was consistent.These showed that the screening about quality trait mutants in M4is effective by this method.3The analysis of farinogram characteristic of Guo9722and Heke2hao showed that the index of water absorption, dough formation time, dough stabilizing time, weakening degree,evaluation value and so on in M5variation strains from two species were different with parents. Variation strains varied on farinogram traits were the mutagenesis offspring226from Guo9722and the mutagenesis offspring242from Heke2. significantly different with parents. wheat variation strains of farinogram traits can obtained by radiation with60Co-rays. 4Use to the protein mark method, the M5generation for high molecular weight glutenin protein (HMW-GS) by electrophoresis analysis. From electrophoresis can be seen, the Guo9722of M5generation are obviously different in HMW-GS bands between11variation lines and parents, The HMW-GS of the parent was5+14+15+9+12,, while that of the variant lines was either1+5+7+9+12or1+5+7+8+12; In Heke2hao,Ms generation have3variation lines to different in HMW-GS bands with parents, The HMW-GS of the parent was2+7+8+9+12, while that of the variant lines was either2+7+8+12. suggesting that variations of glutenin locus can be occurred.
