Research On The Associative Actions Of Fusarium Toxin DON And ZEA On The Function Of Mouse’s Intestinal Tract

We chose180female healthy Kunming mice as samples of our experiment in accordance with the rules of2elements and3standards. The respective consistency of DON is0mg/kgBW、1.5mg/kgBW、2.5mg/kgBW, while the consistency of ZEA is0mg/kgBW、20mg/kgBW、30mg/kgBW. We divided these mice into9groups in random including8for experiment and1for comparison. We named those groups as group of comparison(10%methanol solution)、group Ⅰ(DON1.5mg/kgBW)、group Ⅱ(DON2.5mg/kgBW)、group Ⅲ(ZEA20mg/kgBW)、group Ⅳ(ZEA30mg/kgBW)、group Ⅴ(DON1.5mg/kgBW and ZEA20mg/kgBW)、group Ⅵ(DON1.5mg/kgBW and ZEA30mg/kgBW)、group Ⅶ(DON2.5mg/kgBW and ZEA20mg/kgBW) and group Ⅷ (DON2.5mg/kgBW and ZEA30mg/kgBW). All the mice were continuously injected for4times in abdominal cavity every24hours with different consistency of DON and ZEA. After the first injections, We got4samples respectively from all goups in the3d (before the third injection)、the5d、the8d as well as the12d. We used these samples for dissection and made their upper jejunum into tissue homogenate. According to the data of protein content、activity of digestive ferment、index related to state of oxidation、 Complement3、Interleukin-4and activity of diamine oxidase, which were got from the intestinal tract, we wanted to know the associative actions of fusarium toxin DON and ZEA on the function of mice’s intestinal tract. The conclusions were:(1)In the3d and5d, group Ⅰ、group Ⅱ、group Ⅲ and group IV could lower the content of Total Protein and albumin at all times, that is a dose-effect relationship, group Ⅱ、groupⅢ and group IV were extremely significant different from the group of comparison (P<0.01)。Interaction effect of the associative action of DON and ZEA was so significant in lowering the content of Total Protein and albumin,we called it sub additive effect. That combined group Ⅶ(DON2.5mg/kgBW'ZEA20mg/kgBW) was obviously different from group Ⅱ(DON2.5mg/kgBW) and groupⅢ(ZEA20mg/kgBW)(P<0.01), That combined group Ⅷ(DON2.5mg/kgBW'ZEA30mg/kgBW) was obviously different from group II (DON2.5mg/kgBW) and groupⅣ(ZEA30mg/kgBW)(P<0.01).What’s more important, the combined group Ⅷ with high concentration (DON2.5mg/kg BW and ZEA30mg/kgBW) was the most harmful to the content of Total Protein and albumin. However, those toxin, in the12d, hardly had any effects on the content of Total Protein and albumin.(2) In the3d and5d, group Ⅰ、group Ⅱ、groupⅢ and groupⅣ could lower the activity of SOD, but increase the generation of MDA and the vitality of hydroxy radical at all times, that is a dose-effect relationship, group Ⅰ、group Ⅱ and group IV were extremely significant different from the group for comparison (P<0.01). The associative action of DON and ZEA was so significant in increasing the generation of MDA, lowering the activity of SOD and increasing the vitality of hydroxy radical, we called it sub additive effect.The combined group Ⅷ (DON2.5mg/kgBW and ZEA30mg/kgBW) was obviously different from the group Ⅱ(DON2.5mg/kgBW) and group Ⅳ(ZEA30mg/kgBW). With respect to the oxidative damage, the combined group Ⅵ with high concentration of toxin(DON2.5mg/kgBW and ZEA30mg/kgBW)was the most harmful. However, those toxin, In the8d and12d, hardly had any effects on the oxidative damage of intestinal tract.(3) In the3d and5d, group Ⅰ、group Ⅱ、groupⅢ and group IV could lower the activity of lipase and amylase, which was extremely significant different from the group for comparison.(P<0.01).We called this dose-effect relationship. Interaction effect of the associative action of DON and ZEA was so insignificant in lowering the activity of lipase and amylase that combined group Ⅴ(DON1.5mg/kgBW and ZEA20mg/kgBW)was nearly the same with the group Ⅰ(DON1.5mg/kgBW) and group Ⅲ (ZEA20mg/kgBW)(P>0.05), the combined group Ⅵ(DON1.5mg/kgBW and ZEA30mg/kgBW)was nearly the same with the group Ⅰ(DON1.5mg/kgBW)and group Ⅳ (ZEA30mg/kgBW)(P>0.05). The combined group with high concentration of toxin (DON2.5mg/kg BW and ZEA30mg/kgBW) was the most harmful to the activity of digestive ferment. Those toxin, in the8d and12d, hardly had any effects on the activity of digestive ferment.(4) In the3d and5d, group Ⅰ、group Ⅱ、groupⅢ and group IV could lower both the content of Complement3and the activity of DAO, which was extremely significant different from the group for comparison.(P<0.01).We called this dose-effect relationship. Interaction effect of the associative action of DON and ZEA obviously lowered the content of Complement3and the activity of DAO. We called it sub additive effect. Sometimes, group Ⅵ(DON1.5mg/kgBW and ZEA30mg/kgBW) was totally different from group Ⅰ(DON1.5mg/kgBW) and group Ⅳ(ZEA30mg/kgBW),but sometimes it wasn’t.(P<0.05or P<0.01). Also, combined group Ⅶ (DON2.5mg/kgBW and ZEA20mg/kgBW) sometimes differed from groupⅡ(DON2.5mg/kgBW) and group Ⅲ(ZEA20mg/kgBW), sometimes it didn’t.(P<0.05or P <0.01).what’s more, group Ⅷ(DON2.5mg/kgBW and ZEA30mg/kgBW) sometimes was different from group Ⅱ(DON2.5mg/kgBW) and group IV (ZEA30mg/kgBW),sometimes it wasn’t.(P<0.05or P<0.01)。the combined group with high concentration (DON2.5mg/kgBW and ZEA30mg/kgBW) was the most harmful to the content of Complement3and the activity of DAO. In the3d and5d, group Ⅰ、group Ⅱ、 groupⅢ and group Ⅳcould effect the activity of IL-4, which was extremely significant different from the group for comparison (P<0.01). However, according to different consistency, DON group with low consistency, group Ⅰ(DON1.5mg/kgBW) increased the activity of IL-4, while DON group with high consistency, group Ⅱ,(DON2.5mg/kgBW) inhabited the activity of IL-4. ZEA played a key role in increasing the activity. The interaction of the DON and ZEA did have some effect on the activity of IL-4but no obvious sub additive effect. The combined groupVI (DON1.5mg/kgBW and ZEA30mg/kgBW) could apparently increase the activity of IL-4and had the most effect on it. Those toxin, in the8d and12d, hardly had any effects on the gastrointestinal function. The conclusions drawn from those data above were:fusarium toxin DON and ZEA might do harm to both intestinal mucosa and immune system by oxidative damage so as to disorder the alimentary canal function. Moreover, the associative action of DON and ZEA might make it worse and then destroy the intestinal tract function and break the immunization barrier.
