Ziziphus spinosus Hu,which blong to Zizyphus.Rhamnaceae family, is a commonwild resources in the North of China. It not only has extensive medicinal value, but also hashigh economic value. During the long period of living and evolution, it has been formed astrong adaptability to the environment. Ziziphus spinosus Hu resources are rich in Henanprovince, Ziziphus spinosus Hu germplasm resources with superior traits that have beenscattered in the poor condition of the sites, found by the survey with a long-term and a largenumber of observations. In order to better compare with the main characteristics and economictraits and in-depth study on the value of their identity and comprehensive assessment, the needof collection and preservation of Ziziphus spinosus Hu, germplasm resources and evaluationhad been asked. In this article, the distribution and growth characteristics of Ziziphus spinosusHu, were grasped preliminary by field survey, interview crowd and forestry staff methods,some particular characters of special germplasm resources were also investigated. In order tofind the excellent individuals, fourteen Ziziphus spinosus Hu, samples which from Xinzhengand Xinmi were studied by morphology mark; with leaves of Ziziphus Jujuba and Ziziphusspinosus Hu, as the object of the research, We analysis the chloroplast TRN gene of ZiziphusJujuba and Sour Jujube, The genetic relationship of Ziziphus Jujuba and Ziziphus spinosusHu,has been revealed at molecular level. Some molecular evidences for analysis theinterspecific relationship of the Ziziphus Jujuba were provided, and provide basis for planttaxonomy of the Ziziphus Jujuba. The results and conclusions are listed as the following:1、Ziziphus spinosus Hu, has long been ignored, as a result, which leads to the resources ofZiziphus spinosus Hu,were rapid decreased; the deep Processing ability was insufficient,thecomprehensive utilization level was low and so on.2、Collecting and preserving some particular characters of special germplasm resources ofZiziphus spinosus Hu,by seed propagation,root shoot seedling and grafting.The germplasmresources include: white flowers,big fruit size, two kernel and bitter Ziziphus spinosus Hu, etc.3、 From what has been discussed above, we can have a conclusion, the minimum andmaximum values of fruit weight are0.76g and1.18g respectively. The variable coefficient is15.54%, which is the maximum index of fruit; next is stone weight, which variable coefficientis14.81%. The variable coefficients of kernel vertical length and kernel cross diameter are4.85%and4.36%respectively, which has a little difference and forms the diversity of fruitshape. The minimum variable coefficient of fruit shape index is2.45%. 4、Correlation analysis showed that: fruit weight and edible rate are the highest correlation,while the content of vitamin C and fruit shape are the smaller correlation indicators. Fruitdiameter and the stone index are the smallest correlation indicators. There is an inversecorrelation between titratable acid and most index of Sour jujube, the greater shape, theheavier weight, the sweeter, the smaller acidity and the higher edible rate. The smaller jujubestones have the more content of vitamin C per100grams, edible rate is related to fruit weight,which the heavier weight means the higher edible rate.5、Principal component analysis showed that:the no.9of Xinmi has the best quality,the no.1ofXinzheng has low quality in14samples.6、The analysis test system of gene intergenic spacer sequence on Ziziphus Jujuba.andZiziphus spinosus Hu, was constructed.Each25ul reaction system should contain7.5ultemplate DNA,1ul positive and negative primer,12.5ul Tap,3ul water.The amplificationprogram as follows:predenature at94℃for5min,followed by denature at94℃for1min,annealing at54℃for1min,extend at72℃for1min,35cycles,and the next end at72℃for10min.
