| Aimed to investigate the change of soil microbial diversity after the vegetation restoration and reconstruction in Horqin sandy land,4typical sand-fixation plantations (Caragana microphylla, Hedysarum fruticosum, Artemisia halodendron and Pinus sylvestris var. mongolica) and a moving sandy dune (CK) were selected as the research object. The genetic, the functional and the community structural diversities of soil microorganisms of the different plantations were studied by taking the experimental technologies of molecular biology, biochemistry and analytical chemistry, and the methods of DGGE, Biolog and PLFA respectively.The results show that the plantations in Horqin sandy land are rich in genetic diversity of soil microorganisms, with the order as H. fruticosum’> A. halodendron>CK>P. sylvestris var. mongolica> C. microphylla. There are numerous bacteria in the soil of H. fruticosum plantation resulting in the richness of genetic information and complexity of community structure. The abundance of bacteria in the soil of C. microphylla and P. sylvestris var. mongolica is almost the same as that of H. fruticosum, while the number of species is poorer than others. And the community structure of soil bacteria in CK is uniformity, with the even distribution of species and lack of dominant species, which is different from those in plantations. The common bacteria species in the soil of plantations and CK are Gemmatimonadetes, γ-proteobacterium, Bacillus, Actinobacterium, Pelomonas and β-proteobacterium, while the common bacteria species in the soil of plantations is Sphingomonas. Actinoplanes is the endemic species in the soil of P. sylvestris var. mongolica.The plantations in Horqin sandy land are rich in functional diversity of soil microorganisms, with the order as A. halodendron> P. sylvestris var. mongolica> H. fruticosum> C. microphylla>CK. The soil microbes metabolic activity of the cabon sources as carboxylic acids, amino acids, amines/amides and polymers of P. sylvestris var. mongolica and CK is significant different from other plantations. The diversity of functional soil microorganisms of different plantations mainly embodies in the microbes which metabolize amines/amides, amino acids and carbohydrates.The plantations in Horqin sandy land are rich in community structural diversity of soil microorganisms, with the order as P. sylvestris var. mongolica> C. microphylla> H. fruticosum>A. halodendron>CK. Different plantations have different community structure of soil microbes, while dominant species is similar. There are numerous microbes in the soil of C. microphylla, H. fruticosum and P. sylvestris var. mongolica plantaion than that of A. halodendron. The soil microbe species of P. sylvestris var. mongolica is numerous, and the community structure is complex, relativley. On the contrary, the soil microbe species of A. halodendron is poor, and the community structure is simple.As a whole, the plantations in Horqin sandy land are rich in microbial diversity of soil microorganisms, with the order as H. fruticosum> P. sylvestris var. mongolica> A. halodendron> C. microphylla. |
