The Effects Of Prescriptions Of Traditional Chinese Medicine Yin-nourishing And Yang-tonifying To Expression Of IL-6,TNF-α And PPPAγ In Pcos Rat

This experiment pour the different dosages of Prescriptions of traditional Chinese medicine, Yin-nourishing and Yang-tonifying into PCOS rats artificial model respectively, using radioimmunoassay and ELISA method detection level of serum hormone INS, LH, FSH, PRL, E2, T, P and serum immune cell factor IL-6, TNF-α, at the same time using real-time quantitative PCR to measure expression of the abundance on IL-6, TNF-α, PPAR y mRNA in the rat reproductive axis. Aim to reveal the molecular mechanism of traditional Chinese medicine (Prescriptions of Yin-nourishing and Yang-tonifying) in curing PCOS, by the reproductive axis local gene expression of abundance and metabolic, endocrine, immune mechanism. Providing a reference for clinical treatment PCOS, and also for the inheritance and development of traditional Chinese medicine career, then finding out the theory basis for pioneering accords with modern spectrum of disease treatment path. The results show that:1. Fasting plasma glucose (FPG) and insulin(INS) level of PCOS rat was significantly higher than the blank control group.The Prescriptions of Yin-nourishing and Yang-tonifying may improve the body’s IR condition both,by reducing the PCOS rat’s level of FPG and INS.2. The level of serum hormone LH,PRL,T was significantly higher, while FSH,E2obviously lower than the control group in PCOS rat.The Prescriptions of Yin-nourishing and Yang-tonifying can reduce PCOS rat’s serum level of LH,T,PRL increase FSH,E2so as to improve and maintain endocrine balance for rat.3. PCOS rat’s serum cell factors of IL-6,TNF-α was higher than blank control group, indicates that PCOS rat in state of inflammation.The Prescriptions of Yin-nourishing and Yang-tonifying can reduce PCOS rat’s serum level of immune cell factor IL-6, TNF-α, to ease the inflammatory condition.4. The Prescriptions of Yin-nourishing and Yang-tonifying can significantly downregulate IL-6, TNF-a gene expression of abundance in hypothalamus and ovary. During the experiment, PPARy gene expression of abundance has not seen the change in the rat reproductive axis.
