| Blueberry is becoming more and more popular in the market because of its hearthy and nutrition fruit. The development of blueberry industry was imperatived. However, it could not be put into large-scale development because of its low propagation. Therefore, tissue culture is an important way to solve this problem. This paper studied the optimum essential medium, disinfection measures, optimization vitro culture and rhizogenic system, and transplantation of blueberry. Besides, it also explored the issue of blueberries easy to get brown during the tissue culturing. The results showed as followt:(1) It was the best sterilization method that explanting in75%ethanol for2min and turning to the0.1%corrosive sublimate soaked10min, then washing by sterile water6times. It could reduce pollution to15%, the injury rate of85%, and also the growth of callus and lateral buds well(2) The improved WPM medium was the best for "powder blue" cultivation. It showed the avarage bus was6.2and the fresh weight and dry weight were86.1mg and19.1mg respectively after60days,.(3) Using ZT and NAA promoted the blueberry buds and callus induction effectively. It made the callus appeared earlier, and the shoot differentiation was quicker. It also resulted the the vigorous growth, induction rate higher. The improved ZT2.0+WPM mg/L+NAA0.2mg/L wasthe most suitable blueberry proliferation of growth, and the induction rate was as high as82%.(4) The blueberry subculture choose joined0.2mg/L IBA, activated carbon200-500mg/L and improved1/2WPM medium was the most suitable to rootting,. The treatment of dark training about17d with roots occur, cultivate30d turn light training after60d made observation rooting rate as high as86.9%.(5) It was effectively prevent the explant Browning occurred by useing medium as join Vc liquid and activated carbon content in100mg/L-200mg/L,. And it was effectively prevent callus Browning if join Vc liquid containing carbon and in200mg/L-500mg/L to the the medium To overcome the blueberry generation when early training explant and formation of the early generation appear easily presented the Browning pHenomenon, provides the basis.(6) Put the seedling through exercise after transplanting in the fall. Its root quickly dip by took500mg/LIBA implanted1:1corrosion of fresh and pearlite moss. And it could get more than80%of the survival rate and6cm above average growth of height. |
