Fatty liver is a common metabolic disease for fish. Its characteristics show as hepatic steatosis, ballooning degeneration, etc.when the disease developed,it affected the liver metabolism of the sick fish, decreased the resistance At this time, the invasion of exogenous pathogenic factors often cause mass mortality of the sick fish. Penthorum chinense Pursh was proved has the hepatoprotective effect for human, rat and other vertebrates.However, weather it has the same pharmacological effects in fish, has not been reported. Therefore, this experiment was designed the following three tests to study the pharmacological effects of Penthorum chinense Pursh on grass carp steatohepatitis:1High-fat diet built grass carp fatty liver model This test designed a high fat diet containing8%fat to built grass carp fatty liver. The ballooning, steatosis and hepatic (nuclear) morphology score were used to determine whether the model was successful. During the modeling time, Serum samples were used to detect ALT, AST, TG and CHO level and hepatopancreas samples were used to detect MDA, SOD and CAT level. The results showed that, Six weeks of feeding high-fat (8%) diet made the grass carp built a successful fatty liver model. The change of Pathological parameters pointed that excess fat made the grass carp liver cell vacuolation increased, the lipid droplets dispersed, part of the hepatocyte nuclear enrichment,side-shift,or even necrosis, disintegration, following inflammatory cell infiltration. Biochemical tests results showed that within6weeks grass carp serum AST, ALT, TG and in CHO rise; the hepatopancreas MDA concentration increased, both SOD and CAT activities were from advancing to descenting.2The pharmacological effects of Penthorum chinense Pursh extracts to grass carp fatty liver After6weeks high-fat diet,the model of grass carp were divided into5groups,and were fed with high-fat(model group), Zhibituo(treatment control group), high-dose Penthorum chinense Pursh extracts(2g-kg-1), medium-dose Penthorum chinense Pursh extracts(1g·kg-1) and low-dose Penthorum chinense Pursh extracts(0.5g·kg-1) respectively, another control group was fed with basal diet.The test measuted the hepatopancreas histopathological changes and the biochemical parameters to studied pharmacological effects of Penthorum chinense Pursh extracts against fatty liver of grass carp. The test results showed that:this extracts could reduce t the grass carp liver injury, increase the antioxidant capacity of hepatopancreas and restore liver morphology. The medium-dose (1g·kg-1) represented the best treatment effectiveness. Conparing the model group, the dose significantly promoted hepatocyte self-healing which showed as reduced the grass carp serum ALT and ALT concentration (P<0.05), It also significantly reduced the grass carp serum TG and CHO level (P<0.05).Besides, this dose enhanced the grass carp body’ s antioxidant ability within six weeks, which showed as reducing grass carp hepatopancreas content of MDA(P<0.05) and increasing SOD, CAT activity (P<0.05).3Grass carp ACC1mRNA expression test The samples were from control goup, model group, treatment control gourp and medium-dose group respectively at6th week. The total RNA was extracted by Trizol.Using specific primers, the target gene were gotten by two-step RT-PCR. Semi-quantitative deteced the relative expression of ACC1mRNA in each group. Semi-quantitative RT-PCR analysis showed that continuous high fat diet of grass carp liver ACC1mRNA expression increased, pull root vegetables extracts, the relative gene expression decline.In summary, this study found that feeding high fat diet for six weeks could built fatty liver model of grass carp. The medium-dose Penthorum chinense Pursh extract(lg-kg-1) had a therapeutic effect for the fatty liver model, which reperented as reducing the liver damage and blood lipid content, improving the antioxidant capacity and regulating ACC1mRNA expression.
