In this study, the salmonid samples were collected in the main producing area for identifying of salmon. Research on DNA extraction method for different processing fish products, the results show that the DNA quality and concentration were larger differences of five DNA extraction methods, comprehensive comparison, and the Qiagen kit method was the most optimal for yield and quality, and it was less impact on subsequent PCR amplification. To indentify the Salmonidae and Salmoninae in food, specific primers and probe for real-time PCR were designed according to the conservation region of growth hormone gene of Salmon, results showed that the establishment of real-time PCR method is highly specific for the detection of Salmonidae and Salmoninae, the limit of detection was25pg and100pg, and both of the relative limit of detection were0.01%. The standard curve has higher reaction efficiency, good linear relationship and repeatability. They can be used to rapidly and accurately identify the Salmonidae and Salmoninae ingredient in food. The Salmonidae was identified by barcode technique, and the results were accordance with DNA barcode and Mini barcode. The phylogenetic analysis shows consistency with the morphological classification. The Salmonidae can not be accurately identified to species name in our country as a result of the molecular biology information insufficient.
