With the rapid development of animal husbandry and aquaculture in our country, antibiotics are being used commonly, and the problems brought by antibiotic residues are more serious. In order to solve the contradiction between common using of antibiotics and the negative impacts of the resulting, we have to search, study and develop other products. We will process from enhancing animals’immunity and disease resistance, and reduce the usage of antibiotics gradually, so that antibiotics residues and side effects in animals’body can be reduce.In recent years, through a large number of studies people have found that peptidoglycan can enhance the ability to resist diseases when it is used in animal husbandry and aquaculture and aquatic animals as immunostimulants. However, it is difficult for peptidoglycan of natural synthesis to be used commonly owing to the restriction of the clinical application, because its activity is unstable, purity is not high, it contains impurities and it is easy to produce side effects etc.. On the one hand, the peptidoglycan synthesized by the method of classic carbohydrate chemistry is difficult and costs highly, on the other hand, it just contains16sugar chains. So far, peptidoglycan has not been synthesized through chemical means in the world.Muramyl Dipeptide is the minimum effective structure to show adjuvant activity in bacterial components. In this experiment, we design a reasonable chemical pathway to synthesiza the analogues of Muramyl Dipeptide, and then the analogues pf peptidoglycan is synthesized through click chemistry. The specific process is that with glucosamine hydrochloride as the starting material, through some processes of chemical rezctions like acetylation reaction, halogenating reaction, azide reaction, hydrolysis reaction, acetal reaction, the analogues of Muramyl Dipeptide are synthesized firstly, and then through the catalysis of cuprous ion the analogues of Muramyl Dipeptide are polymerized to synthesize the analogues of peptidoglycan.We study this problem through the angle of bionics, and we hope that the synthesized analogues of peptidoglycan could have the same activity of peptidoglycan. It will be used in livestock production as immunostimulants, and we hope that the using of antibiotics could be reduced effectively.In this experiment, we study and experiment the reaction conditions, time and post processing operations of every steps repeatedly, and the reaction will be improved gradually, but due to time constraints we only get the intermediate product-compound6. Although there is a long distance before we get the analogues of Muramyl Dipeptide, the design of the rational way to synthesize peptidoglycan and the successful starting of this experiment are significant to the whole synthesis experiment, and other researcher can use compound6to continue the study.
