Object:The Ciprofloxacin Hydrochloride (CPLX) sustained-release injection was prepared with the modern pharmacy technique, providing the health care of animals with a prolonged action, safety, and low cost preparation.Materials and Methods:The influential factors of the power of hydrogen, depot preparations and the solvent match were inspected predominantly, and the formation was optimized by orthogonal design test to make the CPLX sustained-release injection. The ultraviolet spectrophotometry was optimized by precision and recovery rate test to inspect the pharmaceutical stability. The term of validity was predicted with Qio methods.The Colibacillus, K.pneumoniae, Streptococcus agalactiae, Staphylococcus aureus and Pseudomonas aeruginosa were taken as the test organisms to inspect the antibacterial activity of CPLX active compound and CPLX sustained-release injection in vitro.The ICR mice and dogs were taken as test animals to inspect the safety of CPLX sustained-release injection through the acute toxicity tests, long-term toxicity test and localized stimulus tests.The goats were taken as test animals to study the pharmacokinetics of drugs.Results:The CPLX sustained-release injection was prepared, and it has a longer half life than the reference demonstrated by the animal experiments. The stability of CPLX sustained-release injection was evaluated by the ultraviolet spectrophotometry established. The validity term of the CPLX sustained-release injection was anticipated with Q10methods is about1.66years. The CPLX suatained-release injection has a higher MIC than the reference, but it has a longer action time than the reference in bacteriostasis effect, which was demonstrated by the bacteriostasis test in vitro. It is safer for mice demonstrated by the acute toxicity test and long-term toxicity test, and the biggest dose is1500mg/kg.It has slight stimulatory for dogs and fits to intramscular injection. It has longer elimination half life time (t1/2β=13.1h) than the reference (t1/2β=6.06h) demonstrtated by the pharmacokinetics test, and the biological availability of the CPLX sustained-release injection is121.62%times to the reference in goats.Conclusion:The CPLX sustained-release injection prepared is safety and effectual with sustained release effect, it has a longer action time, lower cost than reference. Hoping exploit it as a new sustained-release formulation.
