Over the years,most medicinal ginseng rely on the production of artificial cultivation.Carrot (Daucus carota var.sativus Hoffm.) is a kind of health vegetables contain nutrient-rich easy to grow and strong drought waterlogging resistance to disease. Has always been as a model material of Genetics and Cell Engineering. As the artificial cultivation ginseng has long growth cycle which demanding the cultivation conditions and the complexity technical management that makes it difficult to control the quality of cultivated ginseng. For this purpose scientists try them best to research ginseng cell culture over the years. In recent years, Biological technology continues to progress and development to provides a new way as innovation and improvement of germplasm resources. The transfer of the gene or genes and somatic hybridization reproductive isolation between species in nature can be get beyond.This experiment used ginseng and carrot as materia established:callus culture system, embryogenic suspension system. Optimal condition of sprotoplast isolation and protoplast electrofusion and the protoplast culture. Cultivate of ginseng regenerated plants, carrot regenerated plants and callus of protoplast electrofusio products. The experiment result provided the foundation for ginseng and carrot somatic hybridization. Results are as follows:1.The callus of carrot and ginseng was induced, and the embryo callus were induced later. A hormones autotrophic type embryo callus of ginseng was optimized by Box-Behnken experiments.2.The suspension system of embryo callus was established, and a suspension system of embryo ginseng was optimized by Box-Behnken experiments, it can make preparation for isolating protoplast.3.Protoplasts of ginseng and carrot were separated, and a protoplast isolation of a suspension system of embryo ginseng was optimized by Box-Behnken experiments.4.The suspended protoplast of ginseng and carrot was fused and electricity fusion products were obtained. 5.The protoplasts were cultured and regenerated ginseng callus and carrot callus and embryos of ginseng and carrot and regenerated fused callus were obtained.6.Regeneration experiments were proceeded, and we obtained regenerated ginseng and carrot plants and regenerated fused embryos.7.The physical and chemical properties and the active principle of the three types of regenerated callus were compared in this experiments, the ginsenoside of4groups of the fused regenerated callus are all higher than the ginseng regenerated callus. |
