Gloeostereum incarnaturm is edible and pharmic wild epiphyte in our cou-ntry, distributing in mountainous area of China Northeast and Japan Hokkaido. China’s traditional medicine for its early recorded, folk often use its decoction to treat diarrhea in folk, its medicinal value is very considerable. The thesis mainly is studied on how to prepare the high antibacterial activity Gloeostereum incarnaturm’s fermented liquid by optimizing the Gloeostereum incarnaturm fermentation medium and fermentation conditions, research the antibacterial spectrum of Gloeostereum incarnaturm’s fermented liquid and analyze its antibacterial characteristics,Compare the Gloeostereum incarnaturm fermented liquid with the crude Gloeostereum incarnaturm polysaccharides to enhance mice immune. The main research contents are as follows:1.The experiment is studied on how to optimize the fermentation medium and fermentation conditions of Gloeostereum incarnaturm, cultivates the Gloeostereum incarnaturm G105by shake flask.Evaluation is biomass and antibacterial activity.The experiment of antibacterial activity is used on cylinder plate method and the indicator is the Staphylococcus aureus.First the best carbon source and nitrogen source of the fermentation medium is proved that is corn starch and soybean powder by the single factor experiment.Through the uniform design experiment get the fermentation medium of Gloeostereum incarnaturm is get to be:corn starch4.6%, soybean powder1.6%, MgSO40.03%, KH2PO40.03%,K2HPO40.01%, VB11piece/100mL. Then the experiment is studied on the fermentation conditions of Gloeostereum incarnaturm which is the fermentation time, the inoculum size, the liquid volume of shake flask, the initial pH of medium. The conclustion of the experiment is the best fermentation conditions that the fermentation time is7days, the inoculum size is10%, the liquid volume of shake flask is20%, the initial pH of medium is5.0-5.5.2. The experiment is studied on the antibacterial spectrum which is the fermentation liquid of Gloeostereum incarnaturm. The experiment of antibacterial activity is used on cylinder plate method.The indicator is Staphylococcus aureus, Escherichia coli, Salmonella, Bacillus subtilis, Bacillus licheniformis, Bacillus cereus, Saccharomyces, Lactobacillus.The result of the experiment prove the antibacterial spectrum of the fermentation liquid is wide. The fermentation liquid of the Gloeostereum incarnaturm can limit the growth of gram-positive bacteria and gram-negative bacteria, but the effort of gram-positive bacteria is stronger than the effort of gram-negative bacteria. The fermentation liquid of the Gloeostereum incarnaturm is founded that can’t limit the growth of the Saccharomyces. Through studing on the physico-chemical property of the fermentation liquid from heat hardiness and PH tolerance, the Antibacterial substances in the fermentation liquid of the Gloeostereum incarnaturm is founded that have favourable heat hardiness.At the same time it has the favourable acid resistance, but it haven’t antibacterial activity in the alkaline environment and it is inconvertible.3. The experiment is studied on the influence of mice immune with polysaccharide of the Gloeostereum incarnaturm which is separated from the fermentation liquid and mycelium of the Gloeostereum incarnaturm. The influence of mice immune is studied on from the aspects of the immune organ, the non-specific immunity, the cellular immunity, the humoral immunity. The study includes on the experiment which is weight ratio of viscera and body, phagocytosis of macrophage, HC50test, lymphocyte transformation by Con A, flow cytometry. The study is comparesd the efforts of the normal mice immune with the different dosages of the polysaccharide which is separated from the fermentation liquid of the Gloeostereum incarnaturm, the polysaccharide which is separated from the mycelium of the Gloeostereum incarnaturm, the fermentation liquid of the Gloeostereum incarnaturm. The polysaccharide of the fermentation liquid can enhance remarkably the level of the immune organ is founded, the non-specific immunity, the cellular immunity, the humoral immunity. The polysaccharide of the mycelium of the Gloeostereum incarnaturm and the fermentation liquid of the Gloeostereum incarnaturm can enhance remarkably the level ofthe non-specific immunity and the cellular immunity, but the effect of humoral immunity is nonconspicuous.The best dosage of the polysaccharide of the fermentation liquid of the Gloeostereum incarnaturm is5mg/d.
