Endophytic fungi is a big group which living in a period of time within the plant tissue, did not cause obvious disease symptoms of the plant tissue. Endophytic fungi exist widely in plants of nature, recent studies show that endophytic fungi can produce some new natural product, and it can even produce similar or the same compounts with their host plant, thus be concern as a new ways to obtain the resources of medicinal.This selected traditional medicinal plant ginseng (Panax ginseng CA Meyer) as the article of study object, endophytic fungi were isolated from the stems and leaves of the aerial parts of ginseng were purified and identified based on colony morphology and microstructure of the strains. Through the screening of antibacterial activity of the isolates,I determine the purpose strain RL2which isolated from the leaves. Through liquid fermentation of the strain,I got the fermentation products and extract its active ingredients, research the pharmacological activity and chemical composition of the strain.In this study,I isolate46endophytic fungi from the stems and leaves of the ground part of ginseng (Panax ginseng CA Meyer)on the tissue block separation,25was isolated from the stem and21from leaves, based on morphology and significant micro-characterization Alternaria is the dominant species, accounting for43.5%of the total number, Phomopsis accounting for21.7%of the total, the number, type and distribution of endophytic fungi separat from the different parts are different, the number, distribution and population of endophytic fungi in different parts of ginseng are differences.This experiment use E. coli, Bacillus.coli, Staphylococcus aureus, Salmonella enteritidis fecal, S.faecalis, Enterobacter sakazakii,Bacillus subtilis as the test bacteria.The results showed that15strains have antibacterial activity at experimental bacteria of4kinds at least, strains RAS7, RAS9、RB5、RL2、RL3、RL9、RL10、RL21showed varying degrees of inhibitory effect of the test bacteria, especially the strains RL2showed strong antibacterial activity of all test bacteria, the relative inhibitory rate of the ethanol extract of mycelium on S.faecalis, Bacillus coli and Salmonellae enteritidis were54.37%,61.72%and44.84%。Alcohol extracts and the fermentation of strains RL2, broth have the inhibitory effect of six kinds of test bacteria, can be considered has broad spectrum and can be used as the purpose strain.Selection of different medium and appropriate culture conditions on the strain RL2fermentation, filtering out the optimal culture conditions suitable for growth of strains RL2, and conducted a preliminary study on the chemical composition of the fermentation products of strains RL2, Panax ginseng C. A.Meyer polysaccharides and saponins usually have a good pharmacological activity, the experimental extraction of the polysaccharides, saponins. The fermentation of the strain RL2with the same triterpenoid saponins with its host plant, the mycelium also detected steroidal saponins.Antioxidant activity experiments of polysaccharide, the activity of intracellular polysaccharide closer to Vc. Its good antioxidant activity. the effect of increasing concentration gradually increased, showing a dose-effect relationship. When C=1mg/ml, strong scavenging ability of intracellular polysaccharide, the clearance rate was83.3%, its clearance rate was83.21percent of the superoxide anion.This experiment test antibacterial activity of polysaccharides, saponins. The intracellular, extracellular polysaccharides, saponins in the fermentation broth as well as pigment composition have good antibacterial activity.High, and low dose group of intracellular extracellular polysaccharide of strains RL2, compared with the control group were able to enhance the vitality of the mice effectively,and extend the swimming time of mice. After loaded swimming in mice, anatomy of the mouse, remove the liver and blood, detected the amount of liver glycogen, blood lactate, blood urea nitrogen three indicators. The results showed that the high, medium and low dose groups of intracellular, extracellular polysaccharide of the strains RL2, compared ability of three indicators to extend the swimming time with the control group of mice,it can reducing the serum BUN concentration after exercise, increased liver glycogen reserves the amount of enhanced clear lactic acid capacity. High, and low dose group of intracellular, extracellular polysaccharide of strains RL2can improve hypoxia tolerance effectively, and prolonged survival time in anoxic environments, its high, and low dose group compared with the blank group have a very significant difference (p<0.01).
