The function of the wheatpollen-specific gene PSG076is still unknown. The research of its promoter will help us tounderstand the regulation of gene expression during sexual reproduction.Based on the DNA sequence of PSG076promoter, we amplified eight differentlength fragments of PSG076promoter from0.4kb to2.4kb by PCR technology. Then, allthese fragments were cloned into pBI121and using gus as reporter gene. Finally, thesepromoter constructs were transformed into tobacco via Agrobacterium-mediatedtransformation method.PCR tenhnology was used to identify wheat pollen-specific promoter-gus wereinserted into the genome of tobacco by using genomic DNA isolated from regeneratedtransgenic tobacco plants. Gus histochemical staining results demonstrated that the coreregion of this pollen-specific promoter maybe900bp upstream of translation start site.Results obtained in the present study will contribute to analyze the detail of genePSG076and its promoter.
