Carotenoids have antioxidant effects, can strengthen the immune system, increaseendurance. Have a good preventive effect on cancer, cataracts and cardiovasculardisease. Beta-carotene, carotenoids, lutein and zeaxanthin quality of carotenoids inhuman and animal body can convert into vitamin A play the original physiologicalrole of vitamin A. Therefore, PSY and LycB can be introduced into maize byAgrobacterium-mediated transformation. And new maize varieties of high-vitamin Acan be created by transgenic technology, which is of great significance in improvingthe nutritional health of people in poor areas, solving the problem of vitamin Adeficiency and a better use of maize.In this study, there are two parts, one part is using the maize inbred line7922forthe plant material, NB as the basic medium, the size of immature embryos:1-2cm,and the embryonic age:9-11d, through agricultural bacillus-mediated method, theconcentration of Agrobacterium bacteria was OD=0.6~0.8, the infection time was20min, co-culture and resting cultivation time were3d and7d, concentration of ASwas100μmol/L, screening concentration and daub concentration of PPT were8mg/Land200mg/L, ultimately, the transgenic plants was detection by PCR, RT-PCR,further to HPLC analysis, the results display carotenoid content was significantlyhigher than wild-type plants, and in addition to the3rd plant, β-carotene content oftransgenic plants raise the ratio in more than50%. Another part of the experiment, weused the maize inbred line TIANTA5as the plant material. Apical meristem as thereceptor, and disinfection time by0.1%mercuric chloride was10min, vacuuminfecting time is20min, eventually, we obtained65resistant plants by PPT resistancescreening, and the resistance rate was7.89%. Further PCR analysis showed that therewere33positive plants; the total transformation frequency was4.01%.
