With the successful development of the preparation of compound anesthesia for miniature pigs-XFM, in order to provide security for the anesthesia process, and realize the anesthesia time and the controllable degree, the anesthesia research group of the Northeast Agricultural University has developed its corresponding antagonistic agent-miniature pigs-specific narcotic antagonistic agent. Based on the theory of balanced anesthesia and the physiological characteristics of miniature pigs, XFM mixture, a preparation of compound anesthesia for miniature pigs, is developed by the miniature pigs anesthesia research group of the Northeast Agricultural University. This mixture has some characteristics, such as the rapid narcotic induction, longer anesthesia maintenance and smooth recovery etc. However, the promotion and application of any kind of anesthesia drug has to have a corresponding antagonistic agent as security, and to achieve the controllability of anesthesia time with the antagonistic agent. And the anesthetic drugs are central inhibitors, they produce effective sedation, analgesic and muscle relaxant effects, but also restrain the physiological functions of the animal respiratory, circulatory, digestive and other systems. In order to ensure the safety and controllability of the anesthesia drug and regulate anesthesia, regaining and recovery time at any time, through the effects of miniature pigs anesthesia antagonistic agent on neurotransmitters in different brain regions of rats, discuss the regaining mechanism of miniature pigs anesthesia antagonistic agent from the tissue, cellular and Molecular level.SD rats as the research object of this experiment, after the IP XFM, respectively in10min,30min,60min for IP specificity antagonist for miniature pigs’combined anesthetic, using HPLC and ELISA detection method in rat cerebral cortex, cerebellum, thalamus, Hippocampus in brain stem and cerebral tissue fluid in the ACh and content determination of amino acid neurotransmitters concentration, further revealing the mechanism of antagonism of drugs, This specific discussion on small pig regaining function and clinical application of anesthesia antagonistic agent is extremely important. The test results are as follows:1. Reversed-phase liquid chromatography (HPLC) determination contents of four kinds of amino acids in rat brain neurotransmitter:the fast, accurate, simple, high sensitivity and reproducibility in1-200ug/mL good linear relationship, Asp, Glu, Gly and GAB A related coefficients respectively0.9956,0.9968,0.9985,0.9965; coefficient of variation coefficient of variation of days and the day below5%; average recovery rate respectively for95.34%,94.73%,96.37%and95.01%.2. The change of ACh content of the various brain regions in rats:under the action of the specificity antagonist for miniature pigs’combined anesthetic, ACh content significantly elevated in the brain regions, regaining group higher than control group each period.3. The content changes in each brain region of the rats excitability neurotransmitter Asp and Glu: under the action of the specificity antagonist for miniature pigs’combined anesthetic, ACh content significantly elevated in the brain regions, regaining group higher than control group each period.;4. The content changes in rats brain regions inhibitory neurotransmitter Gly and GABA:under the action of the Specificity antagonist for miniature pigs’combined anesthetic, the content of inhibitory neurotransmitter Gly and GABA appears declined, the cerebral cortex in a late regaining group of the linear creep group than in the turn is a small amount of the group appeared increased phenomenon,so two sets of content between Gly have no significant difference.The experimental results show that the effect of neurotransmitter content of the specificity antagonist for miniature pigs’ combined anesthetic on the cerebral cortex, hippocampus, thalamus, cerebellum and brainstem ACh and amino acids is likely to be its one of the important mechanism for regaining. Specificity antagonist for miniature pigs’ combined anesthetic regaining mechanism may be related to increasing content of ACh in the hippocampus and cerebral cortex, cortical, Thalamic, and increasing excitatory neurotransmitter within the brain stem of Asp and Glu content, reducing inhibitory neurotransmitters in the hippocampus and brain stem Gly and GABA content. Experimental study on further mini-pigs-specific narcotic regaining antagonistic agent mechanism with reference value.
