Mycoplasma suis (M. suis) infection was a micro-organism which parasitizes on thesurface of red blood cells and causes anemia, jaundice and fever in swine. Due to the largeeconomic losses M. suis infection bringing to us, the demand is urgent to transform theresearch achievement into practice. Therefore the thread of my research was determined withtaking MSG1protein as the suitable object which had been reasearched more relatively.The MSG1protein was expressed in recombinant E. coli strain, the expressed conditionof MSG1was analysed. Then an ELISA method was built to detect M.suis in clinical samplesbased on the MSG1antigen. Meanwhile, the sensitivity, specificity, coincidence andrepeatability were setted to assess the ELISA method. At last, the MSG1protein wasvaccinated into mice, and the hybridoma cells were obtained. Through screening of thepositive clones, the monoclonal cells were acquired to produce the monoclonal antibodiesagainst MSG1protein.This experiment showed that MSG1protein could expressed abundantly in form ofinclusion bodies in E. coli BL21strains, and purified subsequently. The antigenicity of MSG1was admirable. The ELISA method we built was effective to detect M.suis in clinical sampleswith a infection rate of46.25%, and the positive samples of them can hinder the polyclonalantibody from integrating with the MSG1protein markedly and didn’t react with some otherantigens. The sensitivity and specificity values reached95.92%and97.06%respectively, anda higher detection rate was confirmed through the ELISA method comparing with the IHAmethod. Two monoclonal cells strains that could secrete antibodies stably were acquired byhybridoma technology.The result of this research contributed a supplement to studying MSG1protein furtherly,provided a method to detect M.suis and offered material basis to search a more rapid, moreintuitional, more convenient approach in detecting M.suis infection.
