In order to develop and protect wild Japanese yew (Taxus cuspidata) resource, artificially rapid cultivate and make up for serve market demand shortage, provide new effective solution, we studid the seedling growth conditions and main physiological and biochemical indexes and obtain the optimum fertilization scheme through N、P、K ratio fertilization experiment of four-year-old Japanese yew, in order to provide effective methods and theory basis of nutrient diagnosis and obtain maximum biomass in Japanese yew seedling. We also conducted the water stress experiment on fertilization seedlings, in order to study the response of different N、P、K fertilization ratio seedlings on drought. The results were as follows:The influence of ratio fertilization on height, ground diameter, crown width and chlorophyll content in Japanese yew seedlings was significant. In a certain range, the higher of the N, P, K element content, the faster of each growth index of seedling increases, the more of the chlorophyll content rises, however, the deficiency or excess of N, P, K content both could restrain the growth of the seedlings. Through regression equation fitting on index data of seedlings’ height, ground diameter, crown width and chlorophyll content, we calculated the application content of N, P, K when the relevant indexes were in maximum. As to seedlings’ height, ground diameter, crown diameter, the treatment7was the best fertilization scheme. The seedlings’ height, ground diameter, crown width were respectively47.4cm,0.685cm and43.9cm, and the corresponding application of N, P, K were respectively6g,4g and2g. As to chlorophyll content index, the treatment8was the best fertilization scheme. The chlorophyll content reached6.5524mg·g-1·Fw, and the corresponding application of N, P, K were respectively6g,2g and4g. Conducting the DRIS nutrition diagnosis of Japanese yew seedlings, the result showed that the nutrient of treatment5was in the balance state, the corresponding application of N, P, K were respectively3g,1g and2g.Through conducting the drought stress on the fertilization seedlings, we found that the SOD, POD activity and the MDA content all increased in different degrees. In middle and low N, P, K concentration treatment, SOD and POD activity response positively. The ability of scavenging active oxygen was strong, MDA content increased less. Membrane lipid peroxidation was not serious. In high N, P, K concentration treatment, SOD and POD activity and MDA content instead had opposite performance. This explains the seedlings of high concentrations fertilization treatments have a weak ability of scavenging reactive oxygen, membrane lipid peroxidation was serious. We also could see that the drought resisting ability of the low concentration of N, P, K treatment was higher than the high concentration of N, P, K treatment. This explain water had significant physiological effect on the different fertilizer treatment seedlings. So we should fully consider the combined action of fertilizer application and water in practical applications.
