The truffle is a rare wild edible fungi growing on the ground,and has a high nutritionalvalue.Because of expensive,it is known as the "black diamond" on the table.Yunnan and otherplaces have a small amount of the distribution,But the study of the wild population geneticdiversity is still blank.So far,through the application of EST-SSR molecular markers to study plantand animal genetic diversity is already quite common.However, in the fungus,other species is veryrarely reported in addition to mushrooms.To develop EST-SSRs functional markers in truffle,characterization of simple se-quence repeats(SSRs)were analyzed by7254EST(expressedsequence tag)sequences in Citrus x paradisi from NCBI of7254EST sequences.276non-redundant ESTs of Grapefruit were screened276SSRs were mined out. the mean distancewas1.2kb in non-redundant ESTs.All these SSRs included167kinds of repeat motifs.Amongthem the dinucleotide and hexanucleotide repeats were dominant types,accounting for96.4%.Theproportion of other repeat types are less than0.6%.The CAG, and CAGCAA are the advantages ofthe dinucleotide and hexanucleotide repeats,accounted for dinucleotide and hexanucleotide21.0%and3.6%respectively. This research might lay the foundation for designing the targeted EST-SSRprimers,germplasm,Genetic Map Construction,Comparative mapping and developing EST-SSRmolecular markers by mining the information of EST-SSR loci in truffle EST sequence data.This study attempts to localized online SSR search program websat, developmentenvironment and source code are completely external open source, and explore a new way forChina to develop their own bioinformatics software.
