In year2001,Southern rice black-streaked dwarf disease was first observed in Yangxi county,Jiangyang city,Guangdong Province. Then this disease spreaded rapidly throughout the southern part of China and caused serious yield lose. In year2002,Southern rice black-streaked dwarf disease was first observed in Hanshou county of Hunan Province. In year2009-2010, rice was suffered extensive damage from Southern rice black-streaked dwarf disease in Hunan Province. The nucleotide sequences of genomic segments of the Hainan (HN) and Guangdong (GD) isolates of Southern rice black-streaked dwarf virus(SRBSDV)were determined,however,Hunan isolate was blank. The object of this study is to obtain the nucleotide sequences of genomic segments of Hunan isolate of SRBSDV,and analysis this segments using bioinformatic methods. Followed by a research of the methods to detect the SRBSDV and studied the symptoms of the SRBSDV infected rice along with the host of the SRBSDV. The main results of this study were shown as follow:1) We achieved the nucleotide sequences of genomic segments of Hunan isolate of SRBSDV and compared with the HN and GD isolates of SRBSDV,there were no distinct diversity.2) The study found that SRBSDV SP6protein had leucine zipper structure. Moreover, the glycosylation sites, phosphorylation sites and B cell epitope were predicted by SRBSDV S6,S10amino acid sequences. Studied these proteins expressions and modifications have important reference value.3) Comparing with the previous nested RT-PCR, in this study, we found that the new primer basing on SRBSDV S10and one step RT-PCR method could rapidly detect SRBSDV. The result showed that the new detect SRBSDV method could effectively distinguish the positive and negative samples. The new detect SRBSDV method shortened the testing time and simplified the process, was suitable for massive、rapidly、exactly detection.4) All rice samples with white protruding objects on stems or rarely on leaves in adult stage were positive for SRBSDV. White protruding objects should be unique symptom of SRBSDV infection and evidence of SRBSDV presence. In RRSV-free areas, such as Hunan province, spiral tip, twisted veins, pane-like shrink, or even totally distort leaves even ragged leaf edges should be caused by SRBSDV.5) The results indicated sorghum、wild oat and goosegrass were the new natural hosts of SRBSDV...
