With broader prescription of antibiotics, the drug-resistance effect upon pathogenic bacteria has been more and more compelling. Strains characterized with drug-resistance or multiple drug resistance has emerged with wider spectrum of drugs and shorter duration. China has the largest species of traditional Chinese medicine resources in the world. Chinese traditional medicine has less side effect, it not only cover a wide spectrum of bacteria species but also difficult to trigger drug-resistance effect. Moreover, once being absorbed by human body, it will benefit immune system and help body defends against pathogenic microorganisms. In order to screen for a better drug for treatment of Staphylococcus aureus infection as well as provide a theoretical basis for the industrial extraction of Celastrus root skin. this study integrated various research information, explored multiple ingredients of Celastrus root skin with different extraction methods, compared their efficiency toward Staphylococcus aureus infection. After getting optimum extraction conditions with orthogonal approach, extraction were infected into mice, relative clinical symptoms been carefully recorded and paraffin sections been taken so that pathological alternations been observed.In this study, methods ranging from boiling, ethanol extraction, and ultrasonic extraction have been applied to study the efficiency of Celastrus root skin upon Staphylococcus aureus infection. Results show that ethanol extraction was the best extraction method with18.5mm diameter of average zone of inhibition. Boiling kept the diameter of average zone of inhibition was8.2mm. Ultrasonic extraction remained between those two with10.5mm diameter of average zone of inhibition.With minimum inhibitory and sterilization effect, we got that the optimum ethanol extraction concentration is80%, time48hr and temperature60℃.In the research,paper disk method and microdilution antifungal susceptibility test was applied to compare bacteriostatic effect of the celastrus orbiculatus, Rheum officinale and jasmine. Then multiple methods has been used to analysis bacteriostatic effect,such as excavation method, plating method and method of drug sensitive piece of paper. These results show that South snake cane root skin ethanol-extraction fluid significant increases the sensitivity of staphylococcal aureus to antimicrobial.We injected Staphylococcus aureus into mice of both experiment and control group. Then we injected extraction with optimum condition into experimental group. so that the experiment animal models had been established. After15days, compared to experiment group. mice in control group shown apparent depression and less willingness to walk around. After being sacrificed with bleeding of carotid artery, major organs of control group showed increased size, dark red appearance with congestion. In addition. multiple hemorrhagic lesions had also been observed. Upon microscopic examination, major organs were all with congestion, hemorrhage. and degeneration of parenchymal cells. They were mainly cloudy swelling and hydropic degeneration, blood vessel dilation and macrophage infiltration.These results suggest that Celastrus root skin has a good antibacterial effect toward mice against Staphylococcus aureus under the optimum extraction concentration. time and temperature.
