Objective: The research was conduct to Observe the therapeutic effect of BZP and orPSHC for melamine poisoning medel, in order to provide reference for clinical treatment ofmelamine toxication.Method: In the trail test, the concentration of melamine was500,1000,1500,2000mg/kg and blank group,5male mice each group,30days later, if the crystal wasobserved, the mice can be used as melamine poisoning model.According the mixed level orthogonal test table (L42×29),96male mice was randomlydevided in to16groups, normal saline groupand the treatment groups (2~6). To build thepoisoning model, the mice were administrated0.4mL melamine (1500mg/kg) once everydayfor15and30day successively. The mice was treated by BZP (1.12,0.56,0.28g/mL) and orPSHC (1.44,0.72,0.36g/kg), once everyday, test the serum CRE, BUN and UA at7and15dRespectively, at the end of the treatment, the mice was putted to death and weigh the kidney,caculated the kidney coefficient.Result:(1)The result of the trail test reveal, no mice dead and no crystal was observed in500mg/kg group, no mice dead of1000mg/kg group, only40%can be used as the model, nomice dead of1500mg/kg group,100%can be used as the model, two mice dead of2000mg/kg group, only60%can be used as the model, so the1500mg/kg group was the best doseto build the melamine poisoning model. The histopathology was observed after the modelsuccessfully maked, the kidney of the mice was swelling, color became shallow, and thecapsule was easily pelt off. Coronary cut the kidney tissue found that the color of the kidneycortex was shallow, no obvious changes of medulla. After administrated melamine (1500mg/kg)30d successively, the crystal can be observed and blocked the renal tubule, the wall ofthe renal tubule became thinner, the cells had granular degeneration, necrosis, fall off thetubule, the protein tubule type also exist, haemorrhage between the cortex and medulla of thekidney.(2)After7d treatment, the serum CRE, BUN, and UA of treatment groups (2-16) waslower than that of normal saline group (P<0.05). The serum CRE, BUN and UA after15dtreatment further decreased compared with7d, the serum CRE, BUN of all group waslower in contrast with normal saline group. Except of group5,6and7, all decreasedcompared with normal saline group (P<0.05).The total kidney coefficient indicated, except ofgroup7and9, all have significant difference (P<0.05) compared with normal saline group.The histopathology was observed after the treatment, the kidney of the normal saline wasswelling, color became shallow, Coronary cut the kidney tissue found that the color of thekidney cortex was shallow, no crystal was found in the renal tubule. the wall of the renaltubule became thinner, the cells had granular degeneration, necrosis, fall off the tubule, theprotein tubule type also exist, haemorrhage between the cortex and medulla of the kidney.The kidney color of treatment group was recovery, noobvious change in the kidney. Observed under the microscope, the renal cells were slightly swelling, and slight hemorrhage wasdetected between the cortex and the medulla of some groups, but greatly improved comparedwith control group. The best group was BZP0.28g/mL combined with PSHC1.44g/kg, thekidney was almost recovery.Conclusion:The therapeutic effect of BZP and or PSHC was obvious for mice melamine(1500mg/kg) poison. At15d treatment, BZP0.56g/mL group or PSHC1.44g/kg group wasthe better, At7or15d treatment, BZP0.28g/mL group and PSHC1.44g/kg group was thebetter...
