Based on some rice functional genomic databases such as Rice.Plantbiology. Withthe bioinformatic methods,25genes,which maybe stem/leaf-specific genes, were findout from rice databases.Using semi-quantitative RT-PCR assay, the expression patternof each candidate gene can be understood. Statistica results indicated that there were1rice stem/leaf-specific genes. Here we further studied one Gene named Os01g28500.The RT-PCR analysis showed that Os01g28500gene specifically expressed in ricestem/leaf and glume while did not express in other organs. Real-time PCR analysis alsoindicated that the Os01g28500gene only high expressed in chlorenchyma.With promoter analysis software plantCARE, PLACE. The upstream approximate2kb promoter region of Os01g28500gene was analyzed, there exist significant regulationmotifs which were necessary for chlorenchyma expression.Then5’end and3’end deletion promoter fragments of Os01g28500gene were fusedinto GUS reporter gene and transformed into rice. The preliminary results showed thatOs01g28500promoter was an ideal chlorenchyma specific and strong expressionpromoter.
