In the experiment, using isozyme, gliadin and ISSR,17kinds of agropyron are taken asmaterials to study their genetic diversities from biochemical and molecular aspects. This canprovide scientific basis for germplasm resources preservation and cultivation of fine ofvarieties agropyron varieties. And four agropyrons that in seed stage and seedling stage areused to analyze the physiological indexs. The more drought resistance varieties are got, itprovides theory basis for the breeding and planting agropyron of Heilongjiang Province. Theresulsts are stuted as follows:(1) POD, CAT and EST are analyzed by polyacrylamide gel electrophoresis. It shows27strips on the electrophoresis profile. The appeared rate in more than80%of high frequencybands and30%~80%of intermediate frequency bands each are12, each accounting for44.4%of the total; Low frequency bands is3and accounting for11.2%of the total. It showsthat the genetic variation of agropyron is more abundant. By analyzing cluster,17copies ofagropyron were divided into four classes.(2) Analyzing gliadin of17materials with SDS-polyacrylamide gel electrophoresis,16spectral bands are separated.11of them show polymorphism, the ratio is62.5%. The geneticsimilarity coefficient is from0.60to1,Shannon diversity index evaluation sample geneticdiversity results show that: Average diversity index is0.4872, effective alleles of each site is1.1086. The clustering figure show that,17copies of agropyron is divided into three classes.(3)80bands were amplified with10from35primers, they contained70polymorphicbands and their polymorphism percentage was87.5%; Shannon’s index evaluation indictedthat average Shannon’s index was0.5230and the numbers of effective alleles were1.4426;the genetic similarity coefficient among the17varieties ranged from0.62to0.85; clusteranalysis with UPGMA method showed that the cultivars tested in this study could be dividedinto six groups at the level GS0.68.(4) Germination percentage,Germination potential and drought germination index inseed stage are detected, and physiological index, such as RMC, MDA, SPAD in seeding stageare also measured. We evaluate the drought resistance of the four agropyron. Results showthat the four materials are drought resistant varieties, MengnongNO.1=Bianshui>Menggu-1>Daqingqijia.
